Journal ArticleDOI
Comparative characteristics of mesenchymal stem cells from human bone marrow, adipose tissue, and umbilical cord blood.
Wolfgang Wagner,Frederik Wein,Anja Seckinger,Maria Frankhauser,Ute Wirkner,Ulf Krause,Jonathon Blake,Christian Schwager,Volker Eckstein,Wilhelm Ansorge,Anthony D. Ho +10 more
TL;DR: The results have provided a foundation for a more reproducible and reliable quality control using genotypic analysis for defining MSC.
Journal ArticleDOI
Replicative senescence of mesenchymal stem cells: a continuous and organized process.
Wolfgang Wagner,Patrick Horn,Mirco Castoldi,Anke Diehlmann,Simone Bork,Rainer Saffrich,Vladimir Benes,Jonathon Blake,Stefan M. Pfister,Volker Eckstein,Anthony D. Ho +10 more
TL;DR: The studies indicate that replicative senescence of MSC preparations is a continuous process starting from the first passage onwards, which includes far reaching alterations in phenotype, differentiation potential, global gene expression patterns, and miRNA profiles that need to be considered for therapeutic application of M SC preparations.
Journal ArticleDOI
Genome Sequencing of Pediatric Medulloblastoma Links Catastrophic DNA Rearrangements with TP53 Mutations
Tobias Rausch,David T.W. Jones,Marc Zapatka,Adrian M. Stütz,Thomas Zichner,Joachim Weischenfeldt,Natalie Jäger,Marc Remke,David Shih,Paul A. Northcott,Elke Pfaff,Jelena Tica,Qi Wang,Luca Massimi,Hendrik Witt,Sebastian Bender,Sabrina Pleier,Huriye Cin,Cynthia Hawkins,Christian Beck,Andreas von Deimling,Volkmar Hans,Benedikt Brors,Roland Eils,Wolfram Scheurlen,Jonathon Blake,Vladimir Benes,Andreas E. Kulozik,Olaf Witt,Olaf Witt,Dianna Martin,Cindy Zhang,Rinnat Porat,Diana M. Merino,Jonathan D. Wasserman,Nada Jabado,Adam M. Fontebasso,Lars Bullinger,Frank G. Rücker,Konstanze Döhner,Hartmut Döhner,Jan Koster,Jan J. Molenaar,Rogier Versteeg,Marcel Kool,Uri Tabori,David Malkin,Andrey Korshunov,Michael D. Taylor,Peter Lichter,Stefan M. Pfister,Jan O. Korbel +51 more
TL;DR: The whole-genome sequencing-based analysis of a Sonic-Hedgehog medulloblastoma brain tumor from a patient with a germline TP53 mutation is reported, uncovering massive, complex chromosome rearrangements and connecting p53 status and chromothripsis in specific tumor types.
Journal ArticleDOI
MicroRNA signatures predict oestrogen receptor, progesterone receptor and HER2/ neu receptor status in breast cancer
Aoife Lowery,Nicola Miller,Amanda Devaney,Roisin E McNeill,Pamela A Davoren,Christophe Lemetre,Vladimir Benes,Sabine Schmidt,Jonathon Blake,Graham Ball,Michael J. Kerin +10 more
TL;DR: This study demonstrates that ANN analysis reliably identifies biologically relevant miRNAs associated with specific breast cancer phenotypes and indicates that dysregulated miRNA expression could be a marker for poorer prognosis breast cancer, but that it could also present an attractive target for therapeutic intervention.
Journal ArticleDOI
Aging and replicative senescence have related effects on human stem and progenitor cells.
Wolfgang Wagner,Wolfgang Wagner,Simone Bork,Patrick Horn,Damir Krunic,Thomas Walenda,Anke Diehlmann,Vladimir Benes,Jonathon Blake,Franz Xaver Huber,Volker Eckstein,Petra Boukamp,Anthony D. Ho +12 more
TL;DR: It was striking that several age-related gene expression changes in both MSC and HPC were also differentially expressed upon replicative senescence of MSC in vitro, indicating that the authors' stem and progenitor cells undergo a similar process also in vivo.
