Journal ArticleDOI
Delivery materials for siRNA therapeutics
TL;DR: An introduction to the biological challenges that siRNA delivery materials aim to overcome is provided, as well as a discussion of the way that the most effective and clinically advanced classes of si RNA delivery systems are designed to surmount these challenges.
Journal ArticleDOI
Therapeutic genome editing by combined viral and non-viral delivery of CRISPR system components in vivo
Hao Yin,Chun-Qing Song,Joseph R. Dorkin,Lihua Julie Zhu,Yingxiang Li,Qiongqiong Wu,Angela I. Park,Junghoon Yang,Sneha Suresh,Aizhan Bizhanova,Ankit Gupta,Mehmet Fatih Bolukbasi,Stephen Walsh,Roman L. Bogorad,Guangping Gao,Zhiping Weng,Yizhou Dong,Victor Koteliansky,Victor Koteliansky,Scot A. Wolfe,Robert Langer,Wen Xue,Daniel G. Anderson +22 more
TL;DR: The delivery strategy is applied to a mouse model of human hereditary tyrosinemia and it is shown that the treatment generated fumarylacetoacetate hydrolase (Fah)-positive hepatocytes by correcting the causative Fah-splicing mutation and rescued disease symptoms such as weight loss and liver damage.
Journal ArticleDOI
Synthesis and Biological Evaluation of Ionizable Lipid Materials for the In Vivo Delivery of Messenger RNA to B Lymphocytes
Owen S. Fenton,Kevin J. Kauffman,James C. Kaczmarek,Rebecca L. McClellan,Siddharth Jhunjhunwala,Mark W. Tibbitt,Manhao D. Zeng,Eric A. Appel,Joseph R. Dorkin,Faryal F. Mir,Jung H. Yang,Matthias A. Oberli,Michael W. Heartlein,Frank Derosa,Robert Langer,Daniel G. Anderson +15 more
TL;DR: It is demonstrated that LNP composition alone can be used to modulate the site of protein induction in vivo, highlighting the critical importance of designing and synthesizing new nanomaterials for nucleic acid delivery.
Journal ArticleDOI
Therapeutic efficacy in a hemophilia B model using a biosynthetic mRNA liver depot system.
Frank Derosa,Braydon Charles Guild,Shrirang Karve,Lianne Smith,Kevin T. Love,Joseph R. Dorkin,Kevin J. Kauffman,J Zhang,Barak Yahalom,Daniel G. Anderson,Michael W. Heartlein +10 more
TL;DR: In this paper, nanoparticles formulated with lipids and lipid-like materials were developed for delivery of two separate mRNA transcripts encoding either human erythropoietin (hEPO) or factor IX (hFIX) protein.
Journal Article
Therapeutic efficacy in a hemophilia B model using a biosynthetic mRNA liver depot system
Frank Derosa,Braydon Charles Guild,Shrirang Karve,Lianne Smith,Jiasheng Zhang,Barak Yahalom,M W Heartlein,Kerry R. Love,Joseph R. Dorkin,Kevin J. Kauffman,Daniel G. Anderson +10 more
TL;DR: Therapeutic value was established within a hemophilia B ( FIX knockout (KO) mouse model by demonstrating a marked reduction in Hct loss following injury (incision) to FIX KO mice, and the capacity of producing EPO in non-human primates via delivery of formulated mRNA was demonstrated as elevated EPO protein levels were observed over a 72-h time course.
