Lauffenburger,Galit Alter,Paul Stoffels,Mathai Mammen,Johan Van Hoof,Hanneke Schuitemaker,Dan H. Barouch +77 more
TL;DR: Robust single-shot vaccine protection against SARS-CoV-2 in nonhuman primates is demonstrated and vaccine-elicited neutralizing antibody titers correlated with protective efficacy, suggesting an immune correlate of protection.
Journal ArticleDOI
Ad26 vector-based COVID-19 vaccine encoding a prefusion-stabilized SARS-CoV-2 Spike immunogen induces potent humoral and cellular immune responses.
Rinke Bos,Lucy Rutten,Joan E.M. van der Lubbe,Mark J. G. Bakkers,Gijs Hardenberg,Frank Wegmann,David Zuijdgeest,Adriaan H. de Wilde,Annemart Koornneef,Annemiek Verwilligen,Danielle van Manen,Ted Kwaks,Ronald Vogels,Tim J. Dalebout,Sebenzile K. Myeni,Marjolein Kikkert,Eric J. Snijder,Zhenfeng Li,Dan H. Barouch,Jort Vellinga,Johannes P. M. Langedijk,Roland Zahn,Jerome Custers,Hanneke Schuitemaker +23 more
TL;DR: In vitro characterization demonstrated that the introduction of stabilizing substitutions increased the ratio of neutralizing versus non-neutralizing antibody binding, suggestive for a prefusion conformation of the S protein, and the wild-type signal peptide was best suited for the correct cleavage needed for a natively folded protein.
Journal ArticleDOI
Stabilizing the closed SARS-CoV-2 spike trimer.
Jarek Juraszek,Lucy Rutten,Sven Blokland,Pascale Bouchier,Richard Voorzaat,Tina Ritschel,Mark J. G. Bakkers,Ludovic Renault,Johannes P. M. Langedijk +8 more
TL;DR: In this article, the structure-based design of soluble S trimers with increased yields and stabilities, based on introduction of single point mutations and disulfide-bridges, was reported.

A Universal Approach to Optimize the Folding and Stability of Prefusion-Closed HIV-1 Envelope Trimers

TL;DR: This work reports a universal strategy that provides for correctly folded Env trimers of high quality and yield through a repair-and-stabilize approach, and provides a means to produce prefusion-closed EnV trimers from diverse HIV-1 strains, a substantial advance for vaccine development.
