Abstract: Glioblastoma multiforme (GM) is the most prevalent tumor among gliomas and presents the highest mortality rate among brain tumors. Berberine (BBR) is an alkaloid isoquinoline found in medicinal plants such as Coptis chinensis. Studies have been showed that BBR presents protective activity in mesenchymal cells and neurons, and antitumor properties in breast cancer and hepatocarcinoma. The aim of this study was to investigate the antitumor effects of BBR in GM U87MG cells, as well as to identify, whether such effects are mediated by oxidative stress and canonical apoptotic pathways. After treatment with several concentrations of BBR (10, 25, 100 and 250 µM) for 24, 48 and 72 h of exposure, BBR reduce cell viability of U87MG cells in a concentration- and time-dependent manner. Afterwards, it was observed that BBR, starting at a concentration of 25 µM of 24 h exposure, significantly suppressed proliferation and increased early apoptosis (53.5% ± 11.15 of annexin V+ propidium iodide− cells) compared to untreated cells (7.5% ± 4.6). BBR-induced apoptosis was independent from AMPK activity and did not change total caspase-3 and p-p53 levels. Moreover, BBR (25 μM/24 h) increased oxidative stress in U87MG cells, evidenced by high levels of reactive oxygen species, thiobarbituric acid reactive substance and protein carbonylation. Considering the antitumor effects of BBR in U87MG cells, this compound may be a potential candidate for adjuvant GM treatment.

Abstract: Background: Curcumin has been used as an additive in the diet of animals in recent years due to the potent medicinal properties of this molecule. Objective: To evaluate whether the addition of curcumin to the diet of calves at different phases (pre- and post-weaning) has a positive effect on metabolic profile, performance, and anti-coccidian action. Methods: Thirtythree Holstein calves were selected at various phases of development: Experiment 1 (E1: n=10) 18±7 (pre-weaning), Experiment 2 (E2: n=11) 64±4 (pre-weaning) and Experiment 3 (E3: n=12) 95±8 (post-weaning) days of life. The calves were separated in three groups according to their phase of development. In each experiment, animals were divided into two sub-groups: control and curcumin. The curcumin groups received 200 mg of additive per animal/day either in milk (pre-weaning) or concentrate (post-weaning). Fecal collections were performed on days 0, 10 and 15 of the experiment to count Eimeria oocysts per gram of feces and to perform fecal score analysis. Complete blood counts, oxidant and antioxidant profiles, protein metabolism markers, lipid levels, glucose levels, and animal weights were measured. Analyses of digestibility and composition of the diet used in Experiment 3 (post-weaning) were also performed. Results: Independent of phase, animals that received curcumin had greater weight gain on days 0 to 15 (E1, E2 and E3 p=0.04, 0.001 and 0.001, respectively), probably due to the increased digestibility of hay and concentrate at 72h (p=0.03 and 0.02, respectively). The supplemented calves had lower level of oxidants (thiobarbituric acid reactive substances –TBARS- and reactive oxygen species –ROS-), indicating that free radical levels in serum and lipid peroxidation were lower. This was probably due to increased enzymatic antioxidants gluthatione S-transferase (E1, E2 and E3 p=0.001, 0.001 and 0.02, respectively), catalase (E1 p=0.001) and superoxide dismutase (E3 p=0.001) in treated animals at day 15. Furthermore, calves receiving curcumin had lower numeric number of Eimeria infection during the experimental period, and the difference was significant in day 15 (E1 and E2 p=0.02, and 0.001, respectively). Conclusion: Curcumin supplementation to dairy calves has coccidiostatic potential, favoring weight gain.

Abstract: The central nervous system of the intermediate host plays a central role in lifelong persistence of Toxoplasma gondii as well as the pathogenesis of congenital toxoplasmosis and reactivated infection in immunocompromised individuals. The purinergic system has been implicated in a wide range of immunological pathways for controlling intracellular responses to pathogens, including T. gondii. In the present study, we investigated the effect of resveratrol (RSV) on ectonucleotidases, adenosine deaminase (ADA), and purinergic receptors during chronic infection by T. gondii. For this study, Swiss mice were divided into control (CTL), resveratrol (RSV), infected (INF), and INF+RSV groups. The animals were orally infected with the VEG strain and treated with RSV (100 mg/kg, orally). Ectonucleotidase activities, P2X7, P2Y1, A1, and A2A purinergic receptor density, ROS, and thiobarbituric acid reactive substances levels were measured in the cerebral cortex of mice. T. gondii infection increased NTPDase and reduced ADA activities. Treatment with RSV also affected enzymes hydrolysing extracellular nucleotides and nucleosides. Finally, RSV affected P1 and P2 purinergic receptor expression during T. gondii infection. Overall, RSV-mediated beneficial changes in purinergic signalling and oxidative stress, possibly improving cerebral cortex homeostasis in T. gondii infection.

Abstract: The progression of the human immunodeficiency virus (HIV) infection to acquired immunodeficiency syndrome (AIDS) can be efficiently interrupted by antiretroviral therapy (ART). However, even successfully treated HIV-infected individuals are prone to develop non-AIDS-related diseases that affect the metabolism and several organs and systems. Biomarkers that predict the occurrence of comorbidities may help develop preventive measures. Current research shows that CD4+ T cell counts and viral load do not predict the development of non-AIDS-related diseases. The CD4/CD8 ratio has been indicated as a suitable marker of persistent immune dysfunction and the occurrence of non-AIDS-related events in treated HIV-positive patients. In this study, we explored the relationship between CD4/CD8 ratios, comorbidities, and aging in ART-treated HIV patients on viral suppression. We collected and evaluated data from 352 HIV-positive adults who were virologically suppressed (<40 copies/mL) on ART and with CD4 counts above 350 cells/mm3 . The median age for participants was 46 years, 218 individuals had at least one comorbidity, and 239 had inverted CD4/CD8 ratios (<1). Current CD4/CD8 ratios were predicted by baseline CD4/CD8 ratios and nadir CD4 counts. Despite the high rates of inverted CD4/CD8 ratios and prevalence of comorbidities, no association between them was observed. The prevalence of comorbidities was significantly higher in older individuals, though aging alone did not explain the rate of all individual comorbidities. Low CD4/CD8 ratios were linked to neurocognitive disorders, suggesting that persistent T cell dysfunction contributes to neurocognitive decline.

Abstract: Accumulation of lanthanum oxide nanoparticles (La2O3NPs) in the femur bone of rats after 30 days of oral administration was explored. Also, the influence of La2O3NPs on macro and trace elements in the rats’ femur bone was assessed. Inductively coupled plasma mass spectrometry (ICP-MS) and inductively coupled plasma optical spectrometry (ICP OES) were used for total element determination in the bone after decomposition while laser ablation-ICP-MS (LA-ICP-MS) was used to investigate element distribution (bio-imaging) in the bone. Some differences in element concentrations in the bone between the rats treated with La2O3NPs at 1.0 mg kg−1 (T1), 10.0 mg kg−1 (T2), and 100 mg kg−1 (T3) body weight (bw) and the control rats (CTR) were observed. More differences were observed in the bone of rat treated with 10.0 mg kg−1 La2O3NPs bw. However, the highest change observed was for Mg, which concentration ranged from 5230 ± 12 μg kg−1 for the CTR group to 4130 ± 138 μg kg−1 for the T3 group. Minor changes were observed for Ba, Ca, Cr, Cu, Fe, Mg, Na, Pb, Sr, and Zn between CTR and animals treated with La2O3NPs at the different levels of concentration. It was possible to observe from LA-ICP-MS analysis that La2O3NPs were accumulated only on the surface of the bone, not deeper than about 5 μm. LA-ICP-MS allowed also to investigate the distribution of La and the other elements in a cross section of the femur bone head, where higher amounts of the elements are present at the external part of the bone. Therefore, it was demonstrated that La2O3NPs are incorporated on the surface of the bone and it has a small influence on some of the other elements evaluated.

Abstract: A Aprendizagem Baseada em Problemas (ABP) e uma metodologia de ensino centrada no aluno, promove o pensamento critico, a resolucao de problemas e pode apresentar resultados favoraveis no periodo de ensino remoto, em virtude da suspensao de atividades escolares. A ABP associada a tematica Coronavirus, foi desenvolvida pelo professor da disciplina de Quimica, com 285 estudantes de Ensino Medio, de uma escola publica do municipio de Sao Sepe/RS. Buscou-se avaliar as percepcoes dos estudantes acerca das atividades, por meio do ensino remoto, comparando a ABP em relacao a atividades tradicionais de ensino. Os resultados mostram que o material didatico agrega conhecimento a tematica atual, e juntamente com a metodologia, estimulam o envolvimento dos sujeitos na busca por respostas. A ABP aliada a tematica Coronavirus, permitiu estabelecer novos saberes, o interesse dos estudantes frente as suas atividades escolares, como uma alternativa as metodologias tradicionais.

Abstract: Glutathione (GSH) represents the major nonprotein thiol in cells and, alongside with glutathione-dependent enzymes such as glutathione reductase (GR), glutathione peroxidase (GPx), and glutathione S-transferase (GST), exerts several biological functions including the protection against free radicals and other essential metabolic reactions within the body. Disturbances in the homeostasis of this complex glutathione antioxidant system may damage cells and have been implicated with the development and progression of several human diseases. In this context, the immune and purinergic systems are also essential, since the dysregulation in both systems may also be correlated with numerous diseases. These two networks are closely related and control inflammatory responses, especially by the crosstalk of signaling molecules, receptors, and enzymes; thus, they can exacerbate or slow down the progression of diseases. Based on this background, we aimed to provide a general scenario of the purinergic and immune systems and the connection between both and the modulation of glutathione and glutathione-dependent enzyme expression and activity in the context of health and disease.

Abstract: A utilizacao de recursos didaticos diferenciados para desenvolver tematicas especificas no Ensino de Ciencias pode funcionar como um facilitador no que denominamos como processo de ensino e aprendizagem. Este artigo se propoe a trazer, entao, o uso de uma maquete, utilizada como um recurso didatico para trabalhar uma tematica especifica, sendo pensada e desenvolvida para os Anos Iniciais do Ensino Fundamental. Neste contexto, o objetivo principal da pesquisa foi mostrar a importância do uso de materiais didaticos como maquetes em uma atividade envolvendo a tematica de cuidados preventivos com o grupo das aranhas, visando a discussao e a reflexao deste assunto no Ensino de Ciencias e na Formacao Inicial de Pedagogos/as. A pesquisa desenvolveu-se durante o primeiro semestre de 2019, numa disciplina intitulada ‘Ensino de Ciencias na Educacao Infantil e Anos Iniciais do Ensino Fundamental’, ofertada para academicos/as do curso de Pedagogia. Os dados foram coletados atraves de rodas de conversa e do uso de um Diario de Campo do pesquisador, ambos analisados de forma qualitativa. A utilizacao de uma maquete didatica permitiu aos/as academico/as um melhor aproveitamento do conteudo desenvolvido durante a atividade, de maneira aberta e problematizadora. Alem disso, a maquete utilizada como recurso didatico pode ajudar os/as futuros/as professores/professoras a dinamizar sua aula, promovendo uma maior participacao das criancas durante as discussoes e problematizacoes que irao surgir ao longo da atividade proposta.

Abstract: Contextualizacao: as demandas atuais sociais exigem dos educadores a busca de novos caminhos e novas metodologias de ensino que foquem no protagonismo dos estudantes, favorecam a motivacao e promovam a autonomia destes, buscando pela articulacao do conhecimento teorico-pratico. O objetivo da pesquisa foi desenvolver modelos didaticos durante uma disciplina do curso de Fisioterapia para auxiliar os academicos no estudo de estruturas complexas, visando um maior aprendizado do conteudo teorico desenvolvido em sala de aula. Descricao da Experiencia: o trabalho foi desenvolvido no modulo de Fisioterapia e Saude da Mulher, no periodo de fevereiro a maio de 2018. Os alunos foram divididos em pequenos grupos e as atividades foram desenvolvidas em tres etapas: planejamento, construcao e apresentacao dos modelos didaticos. Cada grupo selecionou o material que foi usado para a confeccao dos modelos. Foi escolhida a criacao de modelos didaticos para promover uma aula mais atrativa e que despertasse maior interesse nos alunos. Impactos: foram confeccionados quatro modelos didaticos os quais representam a anatomia da mama, tipos de mastectomia, o sistema urinario e a gestacao. Pode-se observar que o uso deste tipo de estrategia, permitiu que os alunos experimentassem um processo de construcao do conhecimento, passando por momentos de duvidas e incertezas, mas que estimulou a participacao ativa do estudante, despertando o interesse dos mesmos e permitindo a aproximacao do conhecimento cientifico de maneira interativa e motivadora. Consideracoes finais: verificou-se que o uso de modelos didaticos possibilitou ao discente a troca de concepcoes, a vivencia do trabalho em equipe e da socializacao. O docente conseguiu vivenciar um processo dinâmico, em que fosse o norteador da construcao do conhecimento.

Abstract: espanolLos accidentes causados por animales ponzonosos son un grave problema de salud publica en Brasil. En ese sentido, desarrollar conocimiento sobre esos animales y divulgar informaciones correctas, por medio de actividades educativas, puede ser el camino para disminuir el numero de accidentes y exponer la importancia ecologica de esos animales de interes medico. Esa investigacion tuvo como objetivo principal investigar los conocimientos previos sobre animales ponzonosos de los participantes de un taller realizado en el 6o Seminario Institucional Pibid/ uergs. La investigacion se desarrollo en la Universidad Federal del Rio Grande del Sur, en el municipio de Alegrete/RS – Brasil. Participaron como sujetos de esa investigacion nueve discentes del curso de pedagogia de la uergs y, antes de empezar las actividades del taller, titulado Materiales Didacticos para la Ensenanza de Ciencias, fue aplicado un cuestionario con cinco preguntas objetivas. Despues del analisis cuantitativo de los cuestionarios, se observo que la mayoria de los participantes de taller cometieron errores conceptuales graves sobre la identificacion de los animales ponzonosos, principalmente sobre serpientes y aranas. En ese sentido, se cree que es necesario que el tema de animales ponzonosos sea trabajado en cursos de formacion de profesores de pedagogia. De esa forma, los futuros profesores, ademas del cuidado personal, podran convertirse en multiplicadores de informacion correcta, por medio de ninos y otras personas a su alrededor posibilitando la disminucion del numero de accidentes por medio de la prevencion. EnglishAccidents caused by poisonous animals constitute a serious problem in the public health of Brazil. In this sense, deepening in the knowledge about these animals and diffusing correct information by educational activities could be the path to reduce the number of accidents and display the ecological importance of these animals of medical interest. This research aimed to investigate the previous knowledge about poison animals of the participants of a workshop performed at 6th International Seminar of Pibid/uergs. The research was developed at State University of Rio Grande do Sul, in Alegrete/RS – Brazil. Nine students of uergs pedagogics course participated as subjects of this research and, before starting the activities of the workshop named Didactic Materials for Science Education, a questionnaire composed of five objective questions was applied. After a quantitative analysis of the questionnaires, it was noticed that the majority of the workshop attendees made serious conceptual mistakes about the identification of poisonous animals, mainly about snakes and spiders. In this sense, it is believed that it is necessary to work on the topic of poisonous animals in training courses of pedagogy teachers. Thus future teachers, besides the self-care, can become multipliers of correct information through children and other people around them, which leads to a probable reduction in the number of accidents through prevention. portuguesOs acidentes causados por animais peconhentos constituem um grave problema de saude publica no Brasil. Nesse sentido, acredita-se ser necessario que a tematica animais peconhentos seja trabalhada nos cursos de formacao de professores de pedagogia. Assim, os futuros professores alem do cuidado pessoal, poderao se tornar multiplicadores de informacoes corretas por meio das criancas e outras pessoas de seu convivio, levando a provavel diminuicao do numero de acidentes atraves da prevencao.

Abstract: Os tempos contemporâneos demandam uma educacao que estimule os estudantes a pensar com autonomia e reflexao critica, ao mesmo tempo mudancas na formacao profissional em fisioterapia foram necessarias para que os curriculos atendessem as necessidades especificas das regioes, as expectativas dos docentes e discentes, incluissem a triade ensino, pesquisa e extensao, assim, foram elaboradas as Diretrizes Curriculares Nacionais (DCN). Ainda, as instituicoes de ensino superior (IES) precisam avaliar o clima organizacional que permite conhecer o ambiente de trabalho e o nivel de qualidade praticado nas IES. O objetivo da presente pesquisa foi analisar a percepcao dos discentes de fisioterapia de duas instituicoes, sobre a influencia da implementacao das DCN e aspectos do clima organizacional na sua formacao academica. A presente pesquisa teve uma abordagem quanti-qualitativa. A amostra foi composta por discentes formandos de uma instituicao publica e outra privada que responderam um questionario e os dados foram analisados de forma descritiva. Verificou-se que o curso e os conteudos das disciplinas atenderam parcialmente as expectativas dos discentes, o trabalho em equipe e a relacao com os docentes foram fundamentais para a formacao de qualidade e houveram inovacoes nas metodologias de ensino. A opiniao dos discentes divergiram quanto ao tempo de disciplinas praticas, infraestrutura, atuacao nos tres niveis de atencao e a busca por atividades de promocao, prevencao e reabilitacao, sendo a percepcao dos discentes da instituicao publica de concordarem parcialmente enquanto da instituicao privada concordarem totalmente. Pode-se concluir que implementacao das DCN, bem como o clima organizacional influenciam a formacao academica.

Abstract: O conhecimento em higiene consiste na adocao de medidas de protecao, com o intuito de promover a saude e evitar formas de contaminacao por doencas infectocontagiosas. O objetivo desse estudo foi relatar uma experiencia pedagogica, sobre o uso de uma câmara escura luminescente por meio de uma atividade ludica, visando a discussao e a reflexao da importância de higienizar corretamente as maos para evitar o contagio do Coronavirus e outros micro-organismos infecciosos. Essa atividade foi desenvolvida em uma escola da rede privada, no municipio de Santa Maria/RS, durante o mes de marco de 2020. Os sujeitos dessa atividade foram 26 escolares, pertencentes aos 3° e 4° Anos Iniciais. Foi construida uma câmara escura utilizando uma caixa de papelao e uma lâmpada de luz ultravioleta, alem do gel fluorescente feito com o refil de caneta marca texto misturado com gel de cabelo. Para a coleta de dados utilizou-se o diario de bordo do professor, observacao participante e uma roda de conversa. Os resultados obtidos evidenciam que a câmara escura e uma proposta inovadora no ensino de ciencias, e na prevencao da Covid-19, pois proporcionou a construcao do conhecimento, principalmente quando se objetiva a mudanca de habitos de higiene. Percebeu-se que a utilizacao da câmara escura, como material didatico ludico foi eficaz, pois as criancas tornaram-se multiplicadoras de informacoes corretas sobre a higienizacao das maos ajudando a evitar o contagio do Coronavirus.
