Molecular mechanism of inhibiting the SARS-CoV-2 cell entry facilitator TMPRSS2 with camostat and nafamostat
Tim Hempel,Lluís Raich,Simon Olsson,Simon Olsson,Nurit P. Azouz,Nurit P. Azouz,Andrea M. Klingler,Markus Hoffmann,Stefan Pöhlmann,Marc E. Rothenberg,Frank Noé,Frank Noé +11 more
TL;DR: The binding process of both drugs and a metabolic product of camostat (GBPA) to TMPRSS2, reaching a Michaelis complex (MC) state, which precedes the formation of a long-lived covalent inhibitory state, suggests that nafamostat is more readily available to form the stable covalENT enzyme–substrate intermediate, effectively explaining its high potency.
Journal ArticleDOI
Inhibition of acid sphingomyelinase by ambroxol prevents SARS-CoV-2 entry into epithelial cells.
Alexander Carpinteiro,Barbara Gripp,Markus Hoffmann,Stefan Pöhlmann,Stefan Pöhlmann,Nicolas Hoertel,Michael J. Edwards,Markus Kamler,Johannes Kornhuber,Katrin Anne Becker,Erich Gulbins,Erich Gulbins +11 more
TL;DR: The acid sphingomyelinase/ceramide system has been shown to be important for cellular infection with at least some viruses, for instance, rhinovirus or severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) as mentioned in this paper.

LY6E impairs coronavirus fusion and confers immune control of viral disease
Stephanie Pfaender,Stephanie Pfaender,Katrina B. Mar,Eleftherios Michailidis,Annika Kratzel,Dagny Hirt,Philip V'kovski,Wenchun Fan,Nadine Ebert,Hanspeter Stalder,Hannah Kleine-Weber,Markus Hoffmann,H.-Heinrich Hoffmann,Mohsan Saeed,Mohsan Saeed,Ronald Dijkman,Eike Steinmann,Mary Wight-Carter,Natasha W. Hanners,Stefan Pöhlmann,Tom Gallagher,Daniel Todt,Gert Zimmer,Charles M. Rice,John W. Schoggins,Volker Thiel +25 more
TL;DR: The results demonstrate that LY6E is a critical antiviral immune effector that controls CoV infection and pathogenesis and advance the understanding of immune-mediated control of CoV in vitro and in vivo, knowledge that could help inform strategies to combat infection by emerging CoV.
Journal ArticleDOI
The glycoprotein of vesicular stomatitis virus promotes release of virus-like particles from tetherin-positive cells
Constantin Brinkmann,Markus Hoffmann,Anastasia Lübke,Inga Nehlmeier,Annika Krämer-Kühl,Michael Winkler,Stefan Pöhlmann +6 more
TL;DR: It is shown that the viral glycoprotein (VSV-G) antagonizes tetherin in transfected cells, although with reduced efficiency as compared to the HIV-1 Vpu protein.
