High-throughput gene targeting and phenotyping in zebrafish using CRISPR/Cas9
Gaurav K. Varshney,Wuhong Pei,Matthew C. LaFave,Jennifer Idol,Lisha Xu,Viviana E Gallardo,Blake Carrington,Kevin Bishop,MaryPat Jones,Mingyu Li,Ursula Harper,Sunny C. Huang,Anupam Prakash,Wenbiao Chen,Raman Sood,Johan Ledin,Shawn M. Burgess +16 more
TL;DR: A cloning-free single-guide RNA (sgRNA) synthesis, coupled with streamlined mutant identification methods utilizing fluorescent PCR and multiplexed, high-throughput sequencing is described, which shows that efficiency of CRISPR/Cas9 is sixfold more efficient than other techniques.
Journal ArticleDOI
Killing dental pathogens using antibacterial graphene oxide.
Jianliang He,Xiaodan Zhu,Zhengnan Qi,Chang Wang,Xiaojun Mao,Cailian Zhu,Zhiyan He,Mingyu Li,Zisheng Tang +8 more
TL;DR: GO nanosheets were found to be an effective antibacterial material against dental pathogens and the potential applications in dental care and therapy are promising.
Journal ArticleDOI
Genome sequencing of the perciform fish Larimichthys crocea provides insights into molecular and genetic mechanisms of stress adaptation.
Jingqun Ao,Yinnan Mu,Li-Xin Xiang,Dingding Fan,Mingji Feng,Shicui Zhang,Qiong Shi,Lv-yun Zhu,Ting Li,Yang Ding,Li Nie,Qiuhua Li,Wei-ren Dong,Liang Jiang,Bing Sun,XinHui Zhang,Mingyu Li,Hai-Qi Zhang,ShangBo Xie,Yabing Zhu,Xuanting Jiang,Xianhui Wang,Pengfei Mu,Wei Chen,Zhen Yue,Zhuo Wang,Jun-Jun Wang,Jian-Zhong Shao,Xinhua Chen +28 more
TL;DR: The results reveal the molecular and genetic basis of fish adaptation and response to hypoxia and air exposure, and reveal new aspects of neuro-endocrine-immune/metabolism regulatory networks that may help the fish to avoid cerebral inflammatory injury and maintain energy balance under Hypoxia.
Journal ArticleDOI
Multiplex Conditional Mutagenesis Using Transgenic Expression of Cas9 and sgRNAs
Linlin Yin,Lisette A. Maddison,Mingyu Li,Nergis Kara,Matthew C. LaFave,Gaurav K. Varshney,Shawn M. Burgess,James G. Patton,Wenbiao Chen +8 more
TL;DR: A system for one-generation multiplex conditional mutagenesis in zebrafish using transgenic expression of both cas9 and multiple single guide RNAs and sgRNAs is demonstrated.
Journal ArticleDOI
Interrupted Glucagon Signaling Reveals Hepatic α Cell Axis and Role for L-Glutamine in α Cell Proliferation
E. Danielle Dean,Mingyu Li,Mingyu Li,Nripesh Prasad,Scott Wisniewski,Alison Von Deylen,Jason M. Spaeth,Lisette A. Maddison,Anthony Botros,Leslie R Sedgeman,Nadejda Bozadjieva,Olga Ilkayeva,Anastasia Coldren,Greg Poffenberger,Alena Shostak,Michael C Semich,Kristie Aamodt,Neil Phillips,Hai Yan,Ernesto Bernal-Mizrachi,Jackie D Corbin,Kasey C. Vickers,Shawn Levy,Chunhua Dai,Christopher B. Newgard,Wei Gu,Roland Stein,Wenbiao Chen,Alvin C. Powers +28 more
TL;DR: A hepatic α islet cell axis where glucagon regulates serum AA availability and AAs, especially L-glutamine, regulate α cell proliferation and mass via mTOR-dependent nutrient sensing is indicated.
