Autophagy-based unconventional secretory pathway for extracellular delivery of IL-1β

TL;DR: This work shows that in mammalian cells, autophagy has a hitherto unappreciated positive contribution to the biogenesis and secretion of the proinflammatory cytokine IL‐1β via an export pathway that depends on Atg5, inflammasome, at least one of the two mammalian Golgi reassembly stacking protein (GRASP) paralogues, GRASP55 (GORASP2) and Rab8a.

Dedicated SNAREs and specialized TRIM cargo receptors mediate secretory autophagy.

TL;DR: It is shown that in response to lysosomal damage, the prototypical cytosolic secretory autophagy cargo IL‐1β is recognized by specialized secretoryautosolic cargo receptor TRIM16 and that this receptor interacts with the R‐SNARE Sec22b to recruit cargo to the LC3‐II+ sequestration membranes.

Neutral lipid stores and lipase PNPLA5 contribute to autophagosome biogenesis

TL;DR: It is shown that lipid droplets as cellular stores of neutral lipids including triglycerides contribute to autophagic initiation in a process dependent on PNPLA5, which was needed for optimal initiation of autophagy.
