De Novo CNV Analysis Implicates Specific Abnormalities of Postsynaptic Signalling Complexes in the Pathogenesis of Schizophrenia
George Kirov,Andrew Pocklington,Peter Holmans,Dobril Ivanov,Masashi Ikeda,Douglas M. Ruderfer,Douglas M. Ruderfer,Jennifer L. Moran,Kimberly Chambert,Draga Toncheva,Lyudmila Georgieva,Detelina Grozeva,Marija Fjodorova,Rebecca Louise Wollerton,Elliott Rees,Ivan Nikolov,L N van de Lagemaat,Àlex Bayés,Esperanza Fernández,Pall I. Olason,Yvonne Böttcher,Noboru H. Komiyama,Mark O. Collins,Jyoti S. Choudhary,Kari Stefansson,Hreinn Stefansson,Seth G. N. Grant,Shaun Purcell,Shaun Purcell,Pamela Sklar,Pamela Sklar,Michael Conlon O'Donovan,Michael John Owen +32 more
TL;DR: The data indicate that defects in NMDAR postsynaptic signalling and, possibly, ARC complexes, which are known to be important in synaptic plasticity and cognition, play a significant role in the pathogenesis of schizophrenia.
Journal ArticleDOI
SynGAP Regulates ERK/MAPK Signaling, Synaptic Plasticity, and Learning in the Complex with Postsynaptic Density 95 and NMDA Receptor
Noboru H. Komiyama,Ayako M. Watabe,Holly J. Carlisle,Karen Porter,Paul Charlesworth,Jennifer Monti,Douglas Strathdee,Colin M. O’Carroll,Stephen J. Martin,Richard G. M. Morris,Thomas J. O'Dell,Seth G. N. Grant +11 more
TL;DR: The amount of potentiation induced by stimulation protocols that induce an ERK-independent form of LTP were also significantly reduced in slices fromsynGAP−/+ mice, suggesting that SynGAP may normally regulate Ras isoforms other than H-Ras.
Journal ArticleDOI
Synaptic scaffold evolution generated components of vertebrate cognitive complexity
Jess Nithianantharajah,Jess Nithianantharajah,Noboru H. Komiyama,Noboru H. Komiyama,Andrew McKechanie,Mandy Johnstone,Douglas Blackwood,David St Clair,Richard D. Emes,Louie N. van de Lagemaat,Louie N. van de Lagemaat,Lisa M. Saksida,Timothy J. Bussey,Timothy J. Bussey,Seth G. N. Grant,Seth G. N. Grant +15 more
TL;DR: The first genetic analysis of a vertebrate gene family in cognitive functions measured using computerized touchscreens shows that Dlg-family mutations underlie psychiatric disorders, suggesting that genome evolution expanded the complexity of vertebrate cognition at the cost of susceptibility to mental illness.
Journal ArticleDOI
CYFIP1 coordinates mRNA translation and cytoskeleton remodeling to ensure proper dendritic spine formation
Silvia De Rubeis,Emanuela Pasciuto,Ka Wan Li,Esperanza Fernández,Daniele Di Marino,Andrea Buzzi,Linnaea E. Ostroff,Eric Klann,Fried J. T. Zwartkruis,Noboru H. Komiyama,Seth G. N. Grant,Christel Poujol,Christel Poujol,Daniel Choquet,Daniel Choquet,Tilmann Achsel,Danielle Posthuma,Danielle Posthuma,August B. Smit,Claudia Bagni +19 more
TL;DR: It is shown that brain-derived neurotrophic factor (BDNF)-driven synaptic signaling releases CYFIP1 from the translational inhibitory complex, triggering translation of target mRNAs and shifting CYFip1 into the WAVE regulatory complex.
Journal ArticleDOI
Specific deletion of focal adhesion kinase suppresses tumor formation and blocks malignant progression
Gordon W. McLean,Noboru H. Komiyama,Bryan Serrels,Hidefumi Asano,Louise E. Reynolds,Francesco J. Conti,Kairbaan Hodivala-Dilke,Daniel Metzger,Pierre Chambon,Seth G. N. Grant,Margaret C. Frame +10 more
TL;DR: This work provides the first experimental proof implicating FAK in tumorigenesis, and this is associated with enhanced apoptosis, and it is found no effect on wound re-epithelialization in vivo.
