Inactivating hepatitis C virus in donor lungs using light therapies during normothermic ex vivo lung perfusion
M. Galasso,Jordan J. Feld,Y. Watanabe,M. Pipkin,C. Summers,Aadil Ali,R. Qaqish,Manyin Chen,R. Ribeiro,Khaled Ramadan,Layla Pires,Vanderlei Salvador Bagnato,Cristina Kurachi,Vera Cherepanov,Gray Moonen,A. Gazzalle,Thomas K. Waddell,Mingyao Liu,Shaf Keshavjee,Brian C. Wilson,Atul Humar,Marcelo Cypel +21 more
TL;DR: Physical viral clearance in combination with germicidal light-based therapies during normothermic ex-vivo Lung Perfusion (EVLP), a method for assessment and treatment of injured donor lungs, inactivates HCV virus in a short period of time and is shown to be safe and prevents HCV transmission in the pig model.
Journal ArticleDOI
Prevention of viral transmission during lung transplantation with hepatitis C-viraemic donors: an open-label, single-centre, pilot trial.
Marcelo Cypel,Marcelo Cypel,Jordan J. Feld,M. Galasso,R. Ribeiro,N. Marks,Magdalena Kuczynski,Deepali Kumar,Ilona Bahinskaya,Vanderlei Salvador Bagnato,Cristina Kurachi,Arthur S. Slutsky,Jonathan C. Yeung,Jonathan C. Waddell,Thomas K. Waddell,Shaf Keshavjee,Shaf Keshavjee,Lianne G. Singer,Atul Humar +36 more
TL;DR: This single centre, prospective, open-label, non-randomised trial aimed to assess the safety and efficacy of lung transplantation in humans from HCV-positive donors toHCV-negative recipients after application of ex-vivo lung perfusion (EVLP) plus ultraviolet C (UVC) perfusate irradiation.
Journal ArticleDOI
Short-course, direct-acting antivirals and ezetimibe to prevent HCV infection in recipients of organs from HCV-infected donors: a phase 3, single-centre, open-label study
Jordan J. Feld,Marcelo Cypel,Deepali Kumar,Harel Dahari,R. Ribeiro,N. Marks,Nellie Kamkar,Ilona Bahinskaya,Fernanda Q. Onofrio,Mohamed A Zahoor,Orlando Cerrochi,Kathryn Tinckam,S. Singer,Atul Humar +25 more
TL;DR: This study shows that an ultra-short course of direct-acting antivirals and ezetimibe can prevent the establishment of chronic HCV infection in the recipient, alleviating many of the concerns with transplanting organs from HCV-infected donors.
Journal ArticleDOI
Ex vivo enzymatic treatment converts blood type A donor lungs into universal blood type lungs
Aizhou Wang,R. Ribeiro,Aadil Ameer Ali,E. Brambate,Etienne Abdelnour-Berchtold,Vinicius Schenk Michaelsen,Yu Zhang,Peter Rahfeld,Ha-Rin Moon,H. Gokhale,A. Gazzalle,Prodipto Pal,Mingyao Liu,Thomas K. Waddell,Christine Cserti-Gazdewich,Kathryn Tinckam,Jayachandran N. Kizhakkedathu,Lori J. West,Shaf Keshavjee,Stephen G. Withers,Marcelo Cypel +20 more
TL;DR: Enzymatic removal of blood group A antigens from donor lungs minimizes antibody-mediated injury in a human ex vivo ABO-incompatible transplant model and has the potential to improve fairness in lung allocation for transplantation.
Journal ArticleDOI
Static lung storage at 10°C maintains mitochondrial health and preserves donor organ function.
Aadil Ali,Aizhou Wang,R. Ribeiro,Erika L. Beroncal,Cristina Baciu,M. Galasso,B. Gomes,A. Mariscal,Olivia Hough,Edson Brambate,Etienne Abdelnour-Berchtold,V. Michaelsen,Yu Zhang,A. Gazzalle,Eddy Fan,Laurent Brochard,Laurent Brochard,Jonathan C. Yeung,Thomas K. Waddell,Mingyao Liu,Ana Cristina Andreazza,Shaf Keshavjee,Marcelo Cypel +22 more
TL;DR: In this article, cold static preservation on ice (~4°C) remains the clinical standard of donor organ preservation, however, mitochondrial injury develops during prolonged storage, which limits the extent of time th...
