Journal ArticleDOI
Mouse Heterochromatin Adopts Digital Compaction States without Showing Hallmarks of HP1-Driven Liquid-Liquid Phase Separation
Fabian Erdel,Fabian Erdel,Anne Rademacher,Rifka Vlijm,Jana Tünnermann,Lukas Frank,Robin Weinmann,Elisabeth Schweigert,Klaus Yserentant,Johan Hummert,Caroline Bauer,Sabrina Schumacher,Ahmad Al Alwash,Christophe Normand,Dirk-Peter Herten,Johann Engelhardt,Karsten Rippe +16 more
TL;DR: It is found that heterochromatin foci resemble collapsed polymer globules that are percolated with the same nucleoplasmic liquid as the surrounding euchromatin, which has implications for the understanding of chromatin compartmentalization and its functional consequences.
Journal ArticleDOI
Hydroxylated Fluorescent Dyes for Live-Cell Labeling: Synthesis, Spectra and Super-Resolution STED†
Alexey N. Butkevich,Vladimir N. Belov,Kirill Kolmakov,Viktor V. Sokolov,Heydar Shojaei,Sven C. Sidenstein,Dirk Kamin,Jessica Matthias,Rifka Vlijm,Johann Engelhardt,Stefan W. Hell +10 more
TL;DR: In this article, hydrogenated rhodamines, carbopyronines, carbo-, silico-, and germanorhodamines with absorption maxima in the range of 530-640 µm were used for specific labeling of living cells.
Journal ArticleDOI
STED nanoscopy of the centrosome linker reveals a CEP68-organized, periodic rootletin network anchored to a C-Nap1 ring at centrioles.
Rifka Vlijm,Xue Li,Marko Panic,Diana Rüthnick,Shoji Hata,Frank Herrmannsdörfer,Thomas Kuner,Mike Heilemann,Mike Heilemann,Johann Engelhardt,Johann Engelhardt,Stefan W. Hell,Elmar Schiebel +12 more
TL;DR: The centrosome linker proteins C-Nap1, rootletin, and CEP68 connect the two centrosomes of a cell during interphase into one microtubule-organizing center as mentioned in this paper.
Journal ArticleDOI
Nucleosome Assembly Dynamics Involve Spontaneous Fluctuations in the Handedness of Tetrasomes
TL;DR: These findings reveal unexpected dynamical rearrangements of the nucleosomal structure, suggesting that chromatin can serve as a "twist reservoir," offering a mechanistic explanation for the regulation of DNA supercoiling in chromatin.
Journal ArticleDOI
Dynamics of nucleosomal structures measured by high-speed Atomic Force Microscopy
TL;DR: High-speed atomic force microscopy is used to visualize dynamic processes in nucleosomes and tetrasomes and demonstrates the ability of high speed AFM to characterize protein-DNA interactions.
