Early-onset Amyloid Deposition and Cognitive Deficits in Transgenic Mice Expressing a Double Mutant Form of Amyloid Precursor Protein 695
M. Azhar Chishti,Dun-Shen Yang,Christopher Janus,Amie L. Phinney,Patrick Horne,Jacqueline Pearson,Robert Strome,Noah Zuker,James E. Loukides,Janet French,Sherry Turner,Gianluca Lozza,Mariagrazia Grilli,Suzanne Kunicki,Céline Morissette,Julie Paquette,Francine Gervais,Catherine Bergeron,Paul E. Fraser,George A. Carlson,Peter St George-Hyslop,David Westaway +21 more
TL;DR: High level production of the pathogenic Aβ42 form of Aβ peptide was associated with an early impairment in TgCRND8 mice in acquisition and learning reversal in the reference memory version of the Morris water maze, present by 3 months of age.
Journal ArticleDOI
SOD1 rescues cerebral endothelial dysfunction in mice overexpressing amyloid precursor protein.
Costantino Iadecola,Fangyi Zhang,Kiyoshi Niwa,Chris Eckman,Sherry Turner,Elizabeth R. Fischer,Steven G. Younkin,David R. Borchelt,Karen K. Hsiao,George A. Carlson +9 more
TL;DR: It is found that transgenic mice overexpressing APP have a profound and selective impairment in endothelium–dependent regulation of the neocortical microcirculation, which may contribute to the alterations in cerebral blood flow and to neuronal dysfunction in Alzheimer's dementia.
Journal ArticleDOI
Aβ1–40-related reduction in functional hyperemia in mouse neocortex during somatosensory activation
Kiyoshi Niwa,Linda H. Younkin,Christine Ebeling,Sherry Turner,David Westaway,Steven G. Younkin,Karen H. Ashe,George A. Carlson,Costantino Iadecola +8 more
TL;DR: Transgenic mice overexpressing APP and Abeta have a profound attenuation in the increase in neocortical blood flow elicited by somatosensory activation, which indicates a potentially deleterious mismatch between substrate delivery and energy demands imposed by neural activity.
Journal ArticleDOI
Genetic Modification of the Phenotypes Produced by Amyloid Precursor Protein Overexpression in Transgenic Mice
George A. Carlson,David R. Borchelt,Angela Dake,Sherry Turner,Valerie Danielson,J. Douglas Coffin,Chris Eckman,James Meiners,Steven P. Nilsen,Steven G. Younkin,Karen K. Hsiao +10 more
TL;DR: Differential survival does not appear to reflect genetic differences in APP processing, but rather host responses to APP or its derivatives, and FGF2 transgene overexpression enhances the lethal effects of APP.
Journal ArticleDOI
Structure and polymorphism of the mouse prion protein gene.
David Westaway,Carol M. Cooper,Sherry Turner,M Da Costa,George A. Carlson,Stanley B. Prusiner +5 more
TL;DR: To understand better the physiology and expression of PrPC, mouse PrP gene (Prn-p) was retrieved from yeast artificial chromosome (YAC), cosmid, phage, and cDNA clones to find conserved motifs which may represent binding sites for as yet unidentified transcription factors.
