Defining the features and duration of antibody responses to SARS-CoV-2 infection associated with disease severity and outcome.
Katharina Röltgen,Abigail E. Powell,Oliver F. Wirz,Bryan A. Stevens,Catherine A. Hogan,Javaria Najeeb,Molly Hunter,Hannah Wang,Malaya K. Sahoo,ChunHong Huang,Fumiko Yamamoto,Monali Manohar,Justin Manalac,Ana R. Otrelo-Cardoso,Tho D. Pham,Tho D. Pham,Arjun Rustagi,Angela J. Rogers,Nigam H. Shah,Catherine A. Blish,Jennifer R. Cochran,Theodore S. Jardetzky,James L. Zehnder,Taia T. Wang,Balasubramanian Narasimhan,Saurabh Gombar,Robert Tibshirani,Kari C. Nadeau,Peter S. Kim,Benjamin A. Pinsky,Scott D. Boyd +30 more
TL;DR: Outpatient and asymptomatic individuals’ SARS-CoV-2 antibodies, including IgG, progressively decreased during observation up to five months post-infection, but antibody responses in acute illness were insufficient to predict inpatient outcomes.
Journal ArticleDOI
Proinflammatory IgG Fc structures in patients with severe COVID-19.
Saborni Chakraborty,Joseph C. Gonzalez,Karlie Edwards,Vamsee Mallajosyula,Anthony S. Buzzanco,Robert Sherwood,Cindy Buffone,Nimish Kathale,Susan Providenza,Markus M. Xie,Jason R. Andrews,Catherine A. Blish,Upinder Singh,Haley L. Dugan,Patrick C. Wilson,Tho D. Pham,Scott D. Boyd,Kari C. Nadeau,Benjamin A. Pinsky,Sheng Zhang,Matthew J. Memoli,Jeffery K. Taubenberger,Tasha Morales,Jeffrey M. Schapiro,Gene S. Tan,Gene S. Tan,Prasanna Jagannathan,Taia T. Wang +27 more
TL;DR: It is shown that disease severity in COVID-19 correlates with the presence of proinflammatory IgG Fc structures, including afucosylated IgG1, and Wang and colleagues show that low levels of IgG fucOSylation enhance interactions with activating Fcγ receptors, boosting the inflammatory cytokines associated with severe COVID.
Journal ArticleDOI
Effects of Aging, Cytomegalovirus Infection, and EBV Infection on Human B Cell Repertoires
Chen Wang,Yi Liu,Lan T. Xu,Katherine J. L. Jackson,Krishna M. Roskin,Tho D. Pham,Jonathan Laserson,Eleanor L. Marshall,Katie Seo,Ji-Yeun Lee,David Furman,Daphne Koller,Cornelia L. Dekker,Mark M. Davis,Mark M. Davis,Andrew Fire,Scott D. Boyd +16 more
TL;DR: The findings isolate effects of aging from those of chronic viral infection on B cell repertoires and provide a baseline for understanding human B cell responses to vaccination or infectious stimuli.
Journal ArticleDOI
Human B-cell isotype switching origins of IgE.
Timothy J. Looney,Ji-Yeun Lee,Krishna M. Roskin,Ramona A. Hoh,Jasmine J. King,Jacob Glanville,Yi Liu,Tho D. Pham,Cornelia L. Dekker,Mark M. Davis,Scott D. Boyd +10 more
TL;DR: Evidence is found that secondary isotype switching of mutated IgG1-expressing B cells is the primary source of IgE in human subjects, with lesser contributions from precursors expressing other switched isotypes and rarely IgM or IgD, suggesting that IgE is derived from previously antigen-experienced B-cells rather than naive B cells that typically express low-affinity unmutated antibodies.
Journal ArticleDOI
Normalizing Microbiota-Induced Retinoic Acid Deficiency Stimulates Protective CD8+ T Cell-Mediated Immunity in Colorectal Cancer
Nupur Bhattacharya,Robert Yuan,Tyler R. Prestwood,Hweixian Leong Penny,Michael A. DiMaio,Nathan E. Reticker-Flynn,Charles R. Krois,Justin A. Kenkel,Tho D. Pham,Yaron Carmi,Lorna L. Tolentino,Okmi Choi,Reyna Hulett,Jinshan Wang,Daniel A. Winer,Joseph L. Napoli,Edgar G. Engleman +16 more
TL;DR: Increased colonic expression of the atRA-catabolizing enzyme, CYP26A1, correlated with reduced frequencies of tumoral cytotoxic CD8(+) T cells and with worse disease prognosis in human CRC.
