MicroRNA-9 Controls the Expression of Granuphilin/Slp4 and the Secretory Response of Insulin-producing Cells
Valérie Plaisance,Amar Abderrahmani,Amar Abderrahmani,Véronique Perret-Menoud,Patrick Jacquemin,Frédéric P. Lemaigre,Romano Regazzi +6 more
TL;DR: Evidence is provided that in insulin-producing cells adequate levels of mir-9 are mandatory for maintaining appropriate Granuphilin levels and optimal secretory capacity, and that silencing of Onecut-2 by RNA interference increases Gran Euphilin expression and mimics the effect of Mir-9 on stimulus-induced exocytosis.
Journal ArticleDOI
Endoplasmic Reticulum Stress Links Oxidative Stress to Impaired Pancreatic Beta-Cell Function Caused by Human Oxidized LDL.
Valérie Plaisance,Saška Brajkovic,Saška Brajkovic,Mathie Tenenbaum,Dimitri Favre,Dimitri Favre,Hélène Ezanno,Amélie Bonnefond,Amélie Bonnefond,Caroline Bonner,Valery Gmyr,Julie Kerr-Conte,Benoit R. Gauthier,Christian Widmann,Gérard Waeber,François Pattou,Philippe Froguel,Philippe Froguel,Amar Abderrahmani,Amar Abderrahmani +19 more
TL;DR: The harmful effects of oxidized LDL in beta-cells requires ER stress activation in a manner that involves oxidative stress, and this mechanism may account for impaired beta-cell function in diabetes and can be reversed by antioxidant treatment.
Journal ArticleDOI
Complexin I regulates glucose-induced secretion in pancreatic β-cells
Amar Abderrahmani,Guy Niederhauser,Valérie Plaisance,Marc-Estienne Roehrich,Vincent Lenain,Thierry Coppola,Romano Regazzi,Gérard Waeber +7 more
TL;DR: It is concluded that CPX I plays a critical role in β-cells in the control of the stimulated-exocytosis of insulin.
Journal ArticleDOI
Increased Hepatic PDGF-AA Signaling Mediates Liver Insulin Resistance in Obesity-Associated Type 2 Diabetes.
Amar Abderrahmani,Loic Yengo,Robert Caiazzo,Mickaël Canouil,Stéphane Cauchi,Violeta Raverdy,Valérie Plaisance,Valérie Pawlowski,Stéphane Lobbens,Julie Maillet,Laure Rolland,Raphael Boutry,Gurvan Queniat,Maxime Kwapich,Mathie Tenenbaum,Julien Bricambert,Sophie Saussenthaler,Elodie Anthony,Pooja Jha,Julien Derop,Olivier Sand,Iandry Rabearivelo,Audrey Leloire,Marie Pigeyre,Martine Daujat-Chavanieu,Sabine Gerbal-Chaloin,Tasnim Dayeh,Guillaume Lassailly,Philippe Mathurin,Bart Staels,Johan Auwerx,Annette Schürmann,Catherine Postic,Clemens Schafmayer,Jochen Hampe,Amélie Bonnefond,François Pattou,Philippe Froguel +37 more
TL;DR: Using DNA methylome and transcriptome analyses of livers from obese individuals, this paper found that hypomethylation at a CpG site in PDGFA (encoding platelet-derived growth factor α) and increased PDGF-AA secretion contributes to insulin resistance, opening new therapeutic avenues against Type 2 diabetes and possibly NAFLD.

TL;DR: In the liver of obese patients with T1D, the increased PDGF-AA signaling contributes to insulin resistance, opening new therapeutic avenues against T2D and possibly NAFLD.
