Journal ArticleDOI
PF-06463922 is a potent and selective next-generation ROS1/ALK inhibitor capable of blocking crizotinib-resistant ROS1 mutations
Helen Y. Zou,Qiuhua Li,Lars D. Engstrom,Melissa West,Victoria A. Appleman,Katy A. Wong,Michele McTigue,Ya-Li Deng,Wei Liu,Alexei Brooun,Sergei Timofeevski,Scott Rp McDonnell,Ping Jiang,Matthew D. Falk,Patrick B. Lappin,Timothy Affolter,Timothy C. Nichols,Wenyue Hu,Justine L. Lam,Ted William Johnson,Tod Smeal,Al Charest,Al Charest,Valeria Fantin +23 more
TL;DR: The identification of PF-06463922, a ROS1/anaplastic lymphoma kinase (ALK) inhibitor, with exquisite potency against ROS1 fusion kinases, capable of inhibiting the ROS1G2032R mutation and FIG-ROS1–driven glioblastoma tumor growth in preclinical models is reported.
Journal ArticleDOI
The activity of Gli transcription factors is essential for Kras-induced pancreatic tumorigenesis
Mihir Rajurkar,Wilfredo E. De Jesus-Monge,David R. Driscoll,Victoria A. Appleman,He Huang,Jennifer L. Cotton,David S. Klimstra,Lihua Julie Zhu,Karl Simin,Lan Xu,Andrew P. McMahon,Brian C. Lewis,Junhao Mao +12 more
TL;DR: It is found that transcriptional activation mediated by the Gli family of transcription factors, although dispensable for pancreatic development, is required for Kras-induced proliferation and survival in primary pancreatic epithelial cells in culture and forKras-driven pancreatic intraepithelial neoplasia and PDAC formation in vivo.
Journal ArticleDOI
Activation of WNT/β-Catenin Signaling Enhances Pancreatic Cancer Development and the Malignant Potential Via Up-regulation of Cyr61
Makoto Sano,David R. Driscoll,Wilfredo E. DeJesus-Monge,Brian Quattrochi,Victoria A. Appleman,Jianhong Ou,Lihua Julie Zhu,Nao Yoshida,Shintaro Yamazaki,Tadatoshi Takayama,Masahiko Sugitani,Norimichi Nemoto,David S. Klimstra,Brian C. Lewis +13 more
TL;DR: It is suggested that the WNT/β-catenin signaling pathway enhances pancreatic cancer development and malignancy in part via up-regulation of CYR61.
Journal ArticleDOI
KRAS(G12D)- and BRAF(V600E)-induced transformation of murine pancreatic epithelial cells requires MEK/ERK-stimulated IGF1R signaling.
TL;DR: It is shown that KRASG12D and BRAFV600E enhance PDEC proliferation and increase survival after exposure to apoptotic stimuli in a manner dependent on MEK/ERK and PI3K/AKT signaling, and a novel mechanism of PI3k/AKt activation downstream of activated KRAS is identified.
Journal ArticleDOI
Activation of tumor-cell STING primes NK-cell therapy.
Erik H. Knelson,Elena Ivanova,Mubin Tarannum,Marco Campisi,Patrick H. Lizotte,Matthew A. Barbie +48 more
TL;DR: It is reported that malignant pleural mesothelioma cells robustly express STING and are responsive to STING agonist treatment ex vivo, and the fundamental importance of using human tumor samples to assess innate and cellular immune therapies is revealed.
