A small molecule-kinase interaction map for clinical kinase inhibitors.
Miles A. Fabian,William H. Biggs,Daniel K. Treiber,Corey E. Atteridge,Mihai Azimioara,Mihai Azimioara,Michael G. Benedetti,Michael G. Benedetti,Todd A. Carter,Pietro Ciceri,Philip T. Edeen,Mark Floyd,Julia M. Ford,Margaret Galvin,Jay L Gerlach,Jay L Gerlach,Robert M. Grotzfeld,Sanna Herrgard,Darren E. Insko,Michael A Insko,Andiliy G. Lai,Jean-Michel Lélias,Shamal A. Mehta,Zdravko V. Milanov,Anne Marie Velasco,Lisa M. Wodicka,Hitesh K. Patel,Patrick P. Zarrinkar,David J. Lockhart +28 more
TL;DR: An efficient way to determine kinase inhibitor specificity by measuring binding of small molecules to the ATP site of kinases is described, which represents a systematic small molecule-protein interaction map for clinical compounds across a large number of related proteins.
Journal ArticleDOI
Protein kinase B/Akt-mediated phosphorylation promotes nuclear exclusion of the winged helix transcription factor FKHR1.
TL;DR: It is shown that the activation of phosphatidylinositol 3 (PI3) kinase by extracellular growth factors induces phosphorylation, nuclear export, and transcriptional inactivation of FKHR1, a member of the FK HR subclass of the forkhead family of transcription factors.
Journal ArticleDOI
The Forkhead Transcription Factor Foxo1 Regulates Adipocyte Differentiation
Jun Nakae,Tadahiro Kitamura,Yukari Ido Kitamura,William H. Biggs,Karen C. Arden,Domenico Accili +5 more
TL;DR: It is proposed that Foxo1 plays an important role in the integration of hormone-activated signaling pathways with the complex transcriptional cascade that promotes adipocyte differentiation.
Journal ArticleDOI
Gut Microbiome-Based Metagenomic Signature for Non-invasive Detection of Advanced Fibrosis in Human Nonalcoholic Fatty Liver Disease
Rohit Loomba,Victor Seguritan,Weizhong Li,Tao Long,Niels Klitgord,Archana Bhatt,Parambir S. Dulai,Cyrielle Caussy,Richele Bettencourt,Sarah K. Highlander,Marcus B. Jones,Claude B. Sirlin,Bernd Schnabl,Lauren M. Brinkac,Nicholas J. Schork,Chi-Hua Chen,David A. Brenner,William H. Biggs,Shibu Yooseph,J. Craig Venter,Karen E. Nelson +20 more
TL;DR: Preliminary evidence is provided for a fecal-microbiome-derived metagenomic signature to detect advanced fibrosis in nonalcoholic fatty liver disease.
Journal ArticleDOI
Disruption of forkhead transcription factor (FOXO) family members in mice reveals their functional diversification
Taisuke Hosaka,William H. Biggs,David Tieu,Antonia D. Boyer,Nissi Varki,Webster K. Cavenee,Karen C. Arden +6 more
TL;DR: It is shown that the physiological roles of Foxo genes are functionally diverse in mammals, and their in vivo physiological roles are largely unknown.
