Journal ArticleDOI
PI3Kγ is a molecular switch that controls immune suppression
Megan M. Kaneda,Karen Messer,Natacha Ralainirina,Hongying Li,Christopher J. Leem,Sara Gorjestani,Gyunghwi Woo,Abraham V. Nguyen,Camila C. Figueiredo,Camila C. Figueiredo,Philippe Foubert,Michael C. Schmid,Melissa Pink,David G. Winkler,Matthew Rausch,Vito J. Palombella,Jeffery L. Kutok,Karen McGovern,Kelly A. Frazer,Xuefeng Wu,Michael Karin,Roman Sasik,Ezra E.W. Cohen,Judith A. Varner +23 more
TL;DR: This work demonstrates that therapeutic targeting of intracellular signalling pathways that regulate the switch between macrophage polarization states can control immune suppression in cancer and other disorders.
Journal ArticleDOI
Deubiquitinating enzyme CYLD negatively regulates the ubiquitin-dependent kinase Tak1 and prevents abnormal T cell responses
William Reiley,Wei Jin,Andrew J. Lee,Ato Wright,Xuefeng Wu,Eric F. Tewalt,Timothy O. Leonard,Christopher C. Norbury,Leo R. Fitzpatrick,Minying Zhang,Shao Cong Sun +10 more
TL;DR: It is shown that CYLD plays a pivotal role in regulating T cell activation and homeostasis and prevents spontaneous activation of the Tak1 axis of T cell signaling and maintaining normal T cell function.
Journal ArticleDOI
Regulation of the Deubiquitinating Enzyme CYLD by IκB Kinase Gamma-Dependent Phosphorylation
TL;DR: Evidence that inducible phosphorylation of CYLD is an important mechanism of its regulation is provided and it is suggested that CYLD serves as a novel target of IKK and that the site-specific phosphorylated of CY LD regulates its signaling function.
Journal ArticleDOI
Crystal structure of inhibitor of κB kinase β
Guozhou Xu,Yu Chih Lo,Qiubai Li,Gennaro Napolitano,Xuefeng Wu,Xuliang Jiang,Michel Dreano,Michael Karin,Hao Wu +8 more
TL;DR: Wu et al. as mentioned in this paper reported the crystal structure of IKKβ in complex with an inhibitor, at a resolution of 3.6 A, revealing a trimodular architecture comprising the kinase domain, a ubiquitin-like domain (ULD), and an elongated, α-helical scaffold/dimerization domain (SDD).
Journal ArticleDOI
Regulation of IκB Kinase-related Kinases and Antiviral Responses by Tumor Suppressor CYLD
Minying Zhang,Xuefeng Wu,Andrew J. Lee,Andrew J. Lee,Wei Jin,Mikyoung Chang,Ato Wright,Ato Wright,Tadaatsu Imaizumi,Shao Cong Sun +9 more
TL;DR: It is shown here that the tumor suppressor CYLD has an essential role in preventing aberrant activation of IKKϵ/TBK1, and a role for CYLD in the control of RIG-I ubiquitination is suggested.
