Mitochondrial Patch Clamp of Beige Adipocytes Reveals UCP1-Positive and UCP1-Negative Cells Both Exhibiting Futile Creatine Cycling
Ambre M. Bertholet,Lawrence Kazak,Edward T. Chouchani,Marta G. Bogaczynska,Ishan Paranjpe,Gabrielle L. Wainwright,Alexandre Betourne,Shingo Kajimura,Bruce M. Spiegelman,Yuriy Kirichok +9 more
TL;DR: In this paper, the inner mitochondrial membrane of beige and brown fat was patch clamped to provide a direct comparison of their thermogenic H + leak (I H ), showing that only 15% of inguinal beige adipocytes had I H, while in the rest UCP1-dependent I H was undetectable.
Journal ArticleDOI
The mechanism of MICU-dependent gating of the mitochondrial Ca2+uniporter
Vivek Garg,Vivek Garg,Junji Suzuki,Ishan Paranjpe,Tiffany Unsulangi,Liron Boyman,Lorin S. Milescu,W. Jonathan Lederer,Yuriy Kirichok +8 more
TL;DR: In this paper, the authors provide rigorous analysis of Ca2+ and Na+ fluxes via MCUcx in intact isolated mitochondria to understand the function of MICU subunits.
Book ChapterDOI
Sperm patch-clamp.
Polina V. Lishko,David E. Clapham,David E. Clapham,Betsy Navarro,Betsy Navarro,Yuriy Kirichok +5 more
TL;DR: This chapter covers important aspects of application of the patch-clamp technique to spermatozoa, such as selection of the electrophysiological equipment, isolation of spermutozoa for patch-Clamp experiments, formation of the gigaohm seal with spermatozosa, and transition into the whole-cell mode of recording.
Book ChapterDOI
Patch-Clamp Analysis of the Mitochondrial Calcium Uniporter.
Vivek Garg,Yuriy Kirichok +1 more
TL;DR: This chapter describes how patch-clamp technique can be applied to record the Ca2+ uniporter currents from the mitochondrial inner membrane, isolation of mitochondria from the heart tissue, and preparation of mitoplast using French Press.
Journal ArticleDOI
Synthetic Essentiality of Metabolic Regulator PDHK1 in PTEN-Deficient Cells and Cancers.

Krogan,Asmin Tulpule,Amit J. Sabnis,Trever G. Bivona +38 more
TL;DR: It is reported that the metabolic regulator pyruvate dehydrogenase kinase1 (PDHK1) is a synthetic-essential gene in PTEN-deficient cancer and normal cells.
