Abstract: Usually two factors are not rigidly standardized in the routine tangent screen examination: (1) the stimulus characteristics of the carrier that fixes the spot to the wand, and (2) patient instructions about what aspects of the stimulus they should attend to. Differences in these factors within the range of what is commonly done in the tangent screen test can lead to significant differences in the locations of the isopters. There is also an interesting interaction between these two factors in establishing isopter size.

Abstract: The targeted compounds included Polychlorinated Biphenyls (PCBs), Pesticides (PESTs), Polycyclic Aromatic Hydrocarbons (PAHs) and so on in the Great Lakes Integrated Atmospheric Deposition Network (IADN), which is a platform based on the IoT (Internet of Things) technology to collect environmental pollutants data. While previous studies usually employed traditional statistical approaches to analyze the IADN results, we performed a complete modeling workflow of the total concentrations of PCBs, PESTs, and PAHs (which is referred to as $\sum $ PCBs, $\sum $ PEST s and $\sum $ PAHs orderly) in 1990–2016 samples by using a machine learning algorithm combined with data-driven research method, which lets the model fit the data, so as to change the model to achieve the effect. The main results of this article are as follows, 1) identifying the spatial and temporal trends of POPs (Persistent Organic Pollutants) in the air of the Great Lakes; 2) An appropriate data-driven intelligent model was constructed for the data at EH (Eagle Harbor) and STP(Sturgeon Point) sampling sites, via which we estimated their $\sum $ PCBs, $\sum $ PESTs, and $\sum $ PAHs in the following 4–5 years, showing the concentrations will continue declining with slight fluctuations; 3) The important role which IoT played in smart environmental protection was pointed out.

Abstract: To what degree can the lignin subunits in a monocot be derived from monolignol ferulate (ML-FA) conjugates? This simple question comes with a complex set of variables. Three potential requirements for optimizing ML-FA production are: 1) The presence of an active FERULOYL-CoA MONOLIGNOL TRANSFERASE (FMT) enzyme throughout monolignol production; 2) Suppression or elimination of enzymatic pathways competing for monolignols and intermediates during lignin biosynthesis; and 3) Exclusion of alternative phenolic compounds that participate in lignification. A 16-fold increase in lignin-bound ML-FA incorporation was observed by introducing an AsFMT gene into Brachypodium distachyon. On its own, knocking out the native p-COUMAROYL-CoA MONOLIGNOL TRANSFERASE (BdPMT) pathway that competes for monolignols and the p-coumaroyl-CoA intermediate did not change ML-FA incorporation, nor did partial loss of CINNAMOYL-CoA REDUCTASE1 (CCR1) function, which reduced metabolic flux to monolignols. However, stacking AsFMT into the Bdpmt-1 mutant resulted in a 32-fold increase in ML-FA incorporation into lignin over the wild-type level.

Abstract: Minichromosome maintenance protein (Mcm) 10 is a part of the eukaryotic replication machinery and highly conserved throughout evolution. As a multivalent DNA scaffold, Mcm10 coordinates the action of proteins that are indispensable for lagging strand synthesis, such as the replication clamp, proliferating cell nuclear antigen (PCNA). The binding between Mcm10 and PCNA serves an essential function during DNA elongation and is mediated by the ubiquitination of Mcm10. Here we map lysine 372 as the primary attachment site for ubiquitin on S. cerevisiae Mcm10. Moreover, we identify five additional lysines that can be ubiquitinated. Mutation of lysine 372 to arginine ablates ubiquitination of overexpressed protein and causes sensitivity to the replication inhibitor hydroxyurea in cells that are S-phase checkpoint compromised. Together, these findings reveal the high selectivity of the ubiquitination machinery that targets Mcm10 and that ubiquitination has a role in suppressing replication stress.
