Journal ArticleDOI
Neurotoxic reactive astrocytes are induced by activated microglia
Shane A. Liddelow,Kevin A. Guttenplan,Laura E. Clarke,Frederick C. Bennett,Christopher J. Bohlen,Lucas Schirmer,Mariko L. Bennett,Alexandra E. Münch,Won-Suk Chung,Todd C. Peterson,Daniel K. Wilton,Arnaud Frouin,Brooke A. Napier,Nikhil Panicker,Manoj Kumar,Marion S. Buckwalter,David H. Rowitch,Valina L. Dawson,Ted M. Dawson,Beth Stevens,Ben A. Barres +20 more
TL;DR: It is shown that activated microglia induce A1 astrocytes by secreting Il-1α, TNF and C1q, and that these cytokines together are necessary and sufficient to induce A2 astroCytes, which are abundant in various human neurodegenerative diseases.
Journal ArticleDOI
Normal aging induces A1-like astrocyte reactivity
Laura E. Clarke,Shane A. Liddelow,Chandrani Chakraborty,Alexandra E. Münch,Myriam Heiman,Ben A. Barres +5 more
TL;DR: The aging-induced up-regulation of reactive astrocytes genes was significantly reduced in mice lacking the microglial-secreted cytokines known to induce A1 reactiveAstrocyte formation, indicating that microglia promote astroCyte activation in aging.
Journal ArticleDOI
Neurotoxic reactive astrocytes induce cell death via saturated lipids
Kevin A. Guttenplan,Maya K. Weigel,Priya Prakash,Prageeth R. Wijewardhane,Philip Hasel,Uriel Rufen-Blanchette,Alexandra E. Münch,Jacob A. Blum,Jonathan Fine,Mikaela C. Neal,Kimberley D. Bruce,Aaron D. Gitler,Gaurav Chopra,Shane A. Liddelow,Ben A. Barres +14 more
TL;DR: In this article, instead of a protein, saturated lipids contained in APOE and APOJ lipoparticles mediate astrocyte-induced toxicity by eliminating the formation of long-chain saturated lipid synthesis enzyme ELOVL1.
Journal ArticleDOI
Neurotoxic Reactive Astrocytes Drive Neuronal Death after Retinal Injury
Kevin A. Guttenplan,Benjamin K. Stafford,Rana N. El-Danaf,Drew I. Adler,Alexandra E. Münch,Maya K. Weigel,Andrew D. Huberman,Shane A. Liddelow +7 more
TL;DR: It is demonstrated that the death of RGCs depends on a combination of both an injury to the neurons and the presence of reactive astrocytes, suggesting a model that may explain why reactive astROcytes are toxic only in some circumstances.
Journal ArticleDOI
Hyperglycemia- and neuropathy-induced changes in mitochondria within sensory nerves
Hussein S. Hamid,Colin M. Mervak,Alexandra E. Münch,Nicholas J. Robell,John M. Hayes,Michael T. Porzio,J. Robinson Singleton,A. Gordon Smith,Eva L. Feldman,Stephen I. Lentz +9 more
TL;DR: An in vitro sensory neuron model and an in situ analysis of human intraepidermal nerve fibers from cutaneous biopsies are employed to measure alterations in the size distribution of mitochondria as a result of hyperglycemia and diabetes.
