Smoking induces coordinated DNA methylation and gene expression changes in adipose tissue with consequences for metabolic health
Pei-Chien Tsai,Pei-Chien Tsai,Pei-Chien Tsai,Craig A. Glastonbury,Craig A. Glastonbury,Melissa Eliot,Sailalitha Bollepalli,Idil Yet,Idil Yet,Juan E. Castillo-Fernandez,Elena Carnero-Montoro,Elena Carnero-Montoro,Thomas Hardiman,Tiphaine Martin,Tiphaine Martin,Alice Vickers,Massimo Mangino,Massimo Mangino,Kirsten J. Ward,Kirsi H. Pietiläinen,Panos Deloukas,Panos Deloukas,Tim D. Spector,Ana Viñuela,Eric B. Loucks,Miina Ollikainen,Karl T. Kelsey,Kerrin S. Small,Jordana T. Bell +28 more
TL;DR: The results provide the first comprehensive characterization of coordinated DNA methylation and gene expression markers of smoking in adipose tissue and give insights into understanding the widespread health consequence of smoking outside of the lung.
Journal ArticleDOI
High-throughput micropatterning platform reveals Nodal-dependent bisection of peri-gastrulation-associated versus preneurulation-associated fate patterning.
Mukul Tewary,Dominika Dziedzicka,Joel Ostblom,Laura Prochazka,Nika Shakiba,Tiam Heydari,Daniel Aguilar-Hidalgo,Curtis Woodford,Elia Piccinini,David Becerra-Alonso,Alice Vickers,Blaise Louis,Nafees Rahman,Davide Danovi,Mieke Geens,Fiona M. Watt,Peter W. Zandstra +16 more
TL;DR: In this article, a robust in vitro platform for high-content screening of multiple human pluripotent stem cell (hPSC) lines for their ability to undergo peri-gastrulation-like fate patterning upon bone morphogenetic protein 4 (BMP4) treatment of geometrically confined colonies and observed significant heterogeneity in their differentiation propensities along a gastrulation associable and neuralization associable axis.
Journal ArticleDOI
Spatial profiling of early primate gastrulation in utero
Sophie Bergmann,Christopher A. Penfold,Erin Slatery,Dylan Siriwardena,Charis Drummer,Stephen J. Clark,Stanley E. Strawbridge,Keiko Kishimoto,Alice Vickers,Mukul Tewary,Timo N Kohler,Florian Hollfelder,Wolf Reik,Erika Sasaki,Rüdiger Behr,Thorsten Boroviak +15 more
TL;DR: In this article , spatial transcriptomics and stem-cell-based embryo models were used to reveal the molecular code of lineage specification in the primate embryo and provide an in vivo reference to decipher human development.
Journal ArticleDOI
Identifying Extrinsic versus Intrinsic Drivers of Variation in Cell Behavior in Human iPSC Lines from Healthy Donors.
Alessandra Vigilante,Anna Laddach,Nathalie Moens,Ruta Meleckyte,Andreas Leha,Arsham Ghahramani,Oliver J. Culley,Annie Kathuria,Chloe Hurling,Alice Vickers,Erika Wiseman,Mukul Tewary,Peter W. Zandstra,Richard Durbin,Franca Fraternali,Oliver Stegle,Ewan Birney,Nicholas M. Luscombe,Davide Danovi,Fiona M. Watt +19 more
TL;DR: This study establishes a strategy for examining the genetic basis of inter-individual variability in cell behavior and identifies genes that correlate in expression with intrinsic and extrinsic PEER factors and associate outlier cell behavior with genes containing rare deleterious non-synonymous SNVs.

Pietiläinen,Panos Deloukas,Panos Deloukas,Tim D. Spector,Ana Viñuela,Eric B. Loucks,Miina Ollikainen,Karl T. Kelsey,Kerrin S. Small,Jordana T. Bell +27 more
TL;DR: The results provide the first comprehensive characterization of coordinated DNA methylation and gene expression markers of smoking in adipose tissue, a subset of which link to human cardio-metabolic health and may give insights into the wide ranging risk effects of smoking across the body.
