Journal ArticleDOI
Single-cell dissection of the human brain vasculature
Francisco J. Garcia,Na Sun,Hyeseung Janice Lee,Brianna Godlewski,Hansruedi Mathys,Kyriaki Galani,Blake Zhou,Xueqiao Jiang,Ayesha P. Ng,Julio C. Mantero,Li-Huei Tsai,David A. Bennett,Mustafa Sahin,Manolis Kellis,Myriam Heiman +14 more
TL;DR: This article performed single-cell characterization of the human cerebrovasculature using both ex vivo fresh tissue experimental enrichment and post mortem in silico sorting of human cortical tissue samples.
Journal ArticleDOI
APOE4 impairs myelination via cholesterol dysregulation in oligodendrocytes
Joel W. Blanchard,Leyla Anne Akay,Jose Davila-Velderrain,Djuna von Maydell,Hansruedi Mathys,Shawn M. Davidson,Audrey H. Effenberger,Chih-Yu Chen,Kristal M. Maner-Smith,Ihab Hajjar,Eric A. Ortlund,Michael Bula,Emre Agbas,Ayesha P. Ng,Xueqiao Jiang,Martin Kahn,C Blanco-Duque,Nicolas Lavoie,Liwang Liu,Ricardo Reyes,Yuan-Ta Lin,Tak Ko,Lea R'Bibo,William T. Ralvenius,David A. Bennett,Hugh P. Cam,Manolis Kellis,Li-Huei Tsai +27 more
TL;DR: In this article , a single-cell transcriptomics profiling of post-mortem human brains from APOE4 carriers compared with non-carriers was performed to gain more comprehensive insights into the impact of APOE-4 on the human brain.
Posted ContentDOI
Single-cell multi-region dissection of brain vasculature in Alzheimer’s Disease
Na Sun,Leyla Anne Akay,Mitchell H. Murdock,Yongjin Park,Adele Bubnys,Kyriaki Galani,Hansruedi Mathys,Xueqiao Jiang,Ayesha P. Ng,David A. Bennett,Li-Huei Tsai,Manolis Kellis +11 more
TL;DR: It is shown that APOE4-genotype associated differences are significantly enriched among AD-associated genes in capillary and venule endothelial cells, and subsets of pericytes and fibroblasts, which underlie the vascular dysregulation in APoe4-associated cognitive decline.
Journal ArticleDOI
Single-nucleus multiregion transcriptomic analysis of brain vasculature in Alzheimer’s disease
Na Sun,Leyla Anne Akay,Mitchell H. Murdock,Yongjin Park,Adele Bubnys,Kyriaki Galani,Hansruedi Mathys,Xueqiao Jiang,Ayesha P. Ng,David A. Bennett,Li-Huei Tsai,Manolis Kellis +11 more
Journal ArticleDOI
Lateral mammillary body neurons in mouse brain are disproportionately vulnerable in Alzheimer’s disease
Wen C. Huang,Zhuyu Peng,Mitchell H. Murdock,Liwang Liu,Hansruedi Mathys,Jose Davila-Velderrain,Xueqiao Jiang,Maggie Chen,Ayesha P. Ng,Fatema Abdurrob,Fan Gao,David A. Bennett,Manolis Kellis,Li-Huei Tsai +13 more
TL;DR: Huang et al. as mentioned in this paper used single-cell RNA sequencing of postmortem brain sections of mouse mammillary body and brain slice electrophysiology to show that lateral MB neurons of 5xFAD mice harbored aberrant hyperactivity that correlated with impaired memory in two behavioral tests.
