Journal ArticleDOI
Abscisic Acid Inhibits Type 2C Protein Phosphatases via the PYR/PYL Family of START Proteins
Sang-Youl Park,Pauline Fung,Noriyuki Nishimura,Davin R. Jensen,Hiroaki Fujii,Yang Zhao,Shelley Lumba,Julia Santiago,Americo Rodrigues,Tsz-fung Freeman Chow,Simon E. Alfred,Dario Bonetta,Ruth R. Finkelstein,Nicholas J. Provart,Darrell Desveaux,Pedro L. Rodriguez,Peter McCourt,Jian-Kang Zhu,Julian I. Schroeder,Brian F. Volkman,Sean R. Cutler +20 more
TL;DR: PYR/PYLs are ABA receptors functioning at the apex of a negative regulatory pathway that controls ABA signaling by inhibiting PP2Cs, illustrating the power of the chemical genetic approach for sidestepping genetic redundancy.
Journal ArticleDOI
Two-state allosteric behavior in a single-domain signaling protein.
TL;DR: A strong correlation is found between phosphorylation-driven activation of the signaling protein NtrC and microsecond time-scale backbone dynamics and a dynamic population shift between two preexisting conformations as the underlying mechanism of activation.
Journal ArticleDOI
A gate–latch–lock mechanism for hormone signalling by abscisic acid receptors
Karsten Melcher,L.-M. Ng,L.-M. Ng,X. Edward Zhou,F.-F. Soon,F.-F. Soon,Yong Xu,Kelly Suino-Powell,Sang-Youl Park,Joshua J. Weiner,Hiroaki Fujii,Hiroaki Fujii,Viswanathan Chinnusamy,Viswanathan Chinnusamy,Amanda Kovach,Jun-Jun Li,Jun-Jun Li,Yonghong Wang,Jiayang Li,Francis C. Peterson,Davin R. Jensen,Eu Leong Yong,Brian F. Volkman,Sean R. Cutler,Jian-Kang Zhu,Jian-Kang Zhu,H. Eric Xu +26 more
TL;DR: In this paper, crystal structures of apo and ABA-bound receptors as well as a ternary PYL2-ABA-PP2C complex were reported, showing that apo receptors contain an open ligand-binding pocket flanked by a gate that closes in response to ABA by way of conformational changes in two highly conserved beta-loops.
Journal ArticleDOI
Structural Basis of CXCR4 Sulfotyrosine Recognition by the Chemokine SDF-1/CXCL12
Christopher T. Veldkamp,Christoph Seibert,Francis C. Peterson,Norberto B. de la Cruz,John C. Haugner,Harihar Basnet,Thomas P. Sakmar,Brian F. Volkman +7 more
TL;DR: The nuclear magnetic resonance structure of a constitutively dimeric SDF-1 in complex with a CXCR4 fragment that contains three sulfotyrosine residues important for a high-affinity ligand-receptor interaction is reported.
Journal ArticleDOI
Interconversion between two unrelated protein folds in the lymphotactin native state
Robbyn L. Tuinstra,Francis C. Peterson,Snjezana Kutlesa,E. Sonay Elgin,Michael A. Kron,Brian F. Volkman +5 more
TL;DR: The results demonstrate that the functional repertoire and regulation of a single naturally occurring amino acid sequence can be expanded by access to a set of highly dissimilar native-state structures.
