Microglial activation and tau propagate jointly across Braak stages
Tharick A. Pascoal,Andrea Lessa Benedet,Nicholas J. Ashton,Nicholas J. Ashton,Nicholas J. Ashton,Min Su Kang,Min Su Kang,Joseph Therriault,Mira Chamoun,Melissa Savard,Firoza Z. Lussier,Cecile Tissot,Thomas K. Karikari,Julie Ottoy,Julie Ottoy,Sulantha Mathotaarachchi,Jenna Stevenson,Gassan Massarweh,Michael Schöll,Michael Schöll,Mony J. de Leon,Jean-Paul Soucy,Paul Edison,Kaj Blennow,Kaj Blennow,Henrik Zetterberg,Serge Gauthier,Pedro Rosa-Neto,Pedro Rosa-Neto +28 more
TL;DR: In this paper, the spatial propagation of microglial activation and tau accumulation colocalize in a Braak-like pattern in the living human brain, suggesting an interaction that determines disease progression.
Journal ArticleDOI
Association of Apolipoprotein E ε4 With Medial Temporal Tau Independent of Amyloid-β.
Joseph Therriault,Joseph Therriault,Andrea Lessa Benedet,Andrea Lessa Benedet,Tharick A. Pascoal,Tharick A. Pascoal,Sulantha Mathotaarachchi,Mira Chamoun,Melissa Savard,Emilie Thomas,Min Su Kang,Min Su Kang,Firoza Z. Lussier,Firoza Z. Lussier,Cecile Tissot,Cecile Tissot,Marlee Parsons,Marlee Parsons,Muhammad Naveed Iqbal Qureshi,Paolo Vitali,Gassan Massarweh,Gassan Massarweh,Jean-Paul Soucy,Jean-Paul Soucy,Soham Rej,Paramita Saha-Chaudhuri,Serge Gauthier,Pedro Rosa-Neto +27 more
TL;DR: In both cohorts, APOEε4 was associated in increased tau-PET uptake in the entorhinal cortex and hippocampus independently of amyloid-β, sex, age, and clinical status after multiple comparisons correction, contributing to an evolving framework in which APoeε4 has deleterious consequences in Alzheimer disease beyond its link with amyloids-β.
Journal ArticleDOI
18F-MK-6240 PET for early and late detection of neurofibrillary tangles.
Tharick A. Pascoal,Joseph Therriault,Andrea Lessa Benedet,Melissa Savard,Firoza Z. Lussier,Mira Chamoun,Cecile Tissot,Muhammad Naveed Iqbal Qureshi,Min Su Kang,Sulantha Mathotaarachchi,Jenna Stevenson,Robert Hopewell,Gassan Massarweh,Jean-Paul Soucy,Serge Gauthier,Pedro Rosa-Neto +15 more
TL;DR: The results suggest that the hierarchical six-stage Braak model using 18F-MK-6240 imaging provides an index of early and late tau accumulation as well as disease stage in preclinical and symptomatic individuals.
Journal ArticleDOI
Mild behavioral impairment is associated with β-amyloid but not tau or neurodegeneration in cognitively intact elderly individuals.
Firoza Z. Lussier,Tharick A. Pascoal,Mira Chamoun,Joseph Therriault,Cecile Tissot,Melissa Savard,Min Su Kang,Min Su Kang,Sulantha Mathotaarachchi,Andrea Lessa Benedet,Marlee Parsons,Muhammad Naveed Iqbal Qureshi,Emilie Thomas,Monica Shin,Laurie-Anne Dion,Gassan Massarweh,Gassan Massarweh,Jean-Paul Soucy,Jean-Paul Soucy,I-Huang Tsai,Paolo Vitali,Zahinoor Ismail,Pedro Rosa-Neto,Pedro Rosa-Neto,Serge Gauthier +24 more
TL;DR: This work examines the associations between MBI and Alzheimer's disease (AD) biomarkers in asymptomatic elderly individuals and finds no clear link between these biomarkers and dementia.
Journal ArticleDOI
Determining Amyloid-β Positivity Using 18F-AZD4694 PET Imaging.
Joseph Therriault,Joseph Therriault,Andrea Lessa Benedet,Andrea Lessa Benedet,Tharick A. Pascoal,Tharick A. Pascoal,Melissa Savard,Nicholas J. Ashton,Nicholas J. Ashton,Mira Chamoun,Cecile Tissot,Cecile Tissot,Firoza Z. Lussier,Firoza Z. TL;DR: Good convergence was obtained among several methods for determining an optimal cutoff for 18F-AZD4694 PET positivity, despite conceptual and analytic idiosyncrasies linked with dichotomization of continuous variables.
