Journal ArticleDOI

Mitochondrial transcription factor A regulates mtDNA copy number in mammals

TL;DR: Genetic evidence for a novel role for TFAM in direct regulation of mt DNA copy number in mammals is provided and it is demonstrated that mtDNA copy number is directly proportional to the total TFAM protein levels also in mouse embryos.
Journal ArticleDOI

DNA Replication and Transcription in Mammalian Mitochondria

TL;DR: The mitochondrion was originally a free-living prokaryotic organism, which explains the presence of a compact mammalian mitochondrial DNA in contemporary mammalian cells, and the proposition that some genes required for DNA replication and transcription were acquired together from a phage early in the evolution of the eukaryotic cell.
Journal ArticleDOI

Mitochondrial transcription factors B1 and B2 activate transcription of human mtDNA

TL;DR: The presence of two proteins that interact with mammalian POLRMT may allow flexible regulation of mtDNA gene expression in response to the complex physiological demands of mammalian metabolism.
Journal ArticleDOI

Elongator, a multisubunit component of a novel RNA polymerase II holoenzyme for transcriptional elongation.

TL;DR: Phenotypes of yeast elp1 delta cells demonstrated an involvement of elongator in transcriptional elongation as well as activation in vivo, and data indicate that the transition from transcriptional initiation to elongation involves an exchange of the multiprotein mediator complex for elongators in a reaction coupled to CTD hyperphosphorylation.
