Journal ArticleDOI
MEG3 long noncoding RNA regulates the TGF-β pathway genes through formation of RNA–DNA triplex structures
Tanmoy Mondal,Santhilal Subhash,Roshan Vaid,Stefan Enroth,Sireesha Uday,Björn Reinius,Sanhita Mitra,Arif Mohammed,Alva Rani James,Emily Hoberg,Aristidis Moustakas,Ulf Gyllensten,Steven J.M. Jones,Claes M. Gustafsson,Andrew H. Sims,Fredrik Westerlund,Eduardo Gorab,Chandrasekhar Kanduri +17 more
TL;DR: It is shown that MEG3 and EZH2 share common target genes, including the TGF-β pathway genes, and RNA–DNA triplex formation could be a general characteristic of target gene recognition by the chromatin-interacting lncRNAs.
Journal ArticleDOI
Human Tumor-Associated Macrophage and Monocyte Transcriptional Landscapes Reveal Cancer-Specific Reprogramming, Biomarkers, and Therapeutic Targets.
Luca Cassetta,Stamatina Fragkogianni,Andrew H. Sims,Agnieszka Swierczak,Lesley M. Forrester,Hui Zhang,Daniel Y.H. Soong,Tiziana Cotechini,Pavana Anur,Elaine Y. Lin,Antonella Fidanza,Martha Lopez-Yrigoyen,Michael Millar,Alexandra Urman,Zhichao Ai,Paul T. Spellman,E. Shelley Hwang,J Michael Dixon,Lisa Wiechmann,Lisa M. Coussens,Harriet O. Smith,Jeffrey W. Pollard,Jeffrey W. Pollard +22 more
TL;DR: It is shown that monocyte subpopulation distribution and transcriptomes are significantly altered by the presence of endometrial and breast cancer and an auto-regulatory loop between TAMs and cancer cells driven by tumor necrosis factor alpha involving SIGLEC1 and CCL8 is identified.
Journal ArticleDOI
Nuclear FAK Controls Chemokine Transcription, Tregs, and Evasion of Anti-tumor Immunity
Alan Serrels,Tom Lund,Bryan Serrels,Adam Byron,Rhoanne C. McPherson,Alex von Kriegsheim,Laura Gómez-Cuadrado,Marta Canel,Morwenna Muir,Jennifer E. Ring,Eleni Maniati,Andrew H. Sims,Jonathan A. Pachter,Valerie G. Brunton,Nick Gilbert,Stephen M. Anderton,Robert J. B. Nibbs,Margaret C. Frame +17 more
TL;DR: Focal adhesion kinase inhibitors may trigger immune-mediated tumor regression, providing previously unrecognized therapeutic opportunities in cancerous squamous epithelial cells.
Journal ArticleDOI
Origins of breast cancer subtypes and therapeutic implications
TL;DR: The common distinguishing features of these breast cancer subtypes are described and it is explained how these features relate both to prognosis and to selection of the most appropriate therapy.
Journal ArticleDOI
Tissue of origin determines cancer-associated CpG island promoter hypermethylation patterns
Duncan Sproul,Robert R. Kitchen,Robert R. Kitchen,Colm E. Nestor,J Michael Dixon,Andrew H. Sims,David J. Harrison,David J. Harrison,Bernard H Ramsahoye,Richard R. Meehan +9 more
TL;DR: The methylation profiles of 1,154 cancers from 7 different tissue types are analyzed and it is suggested that aberrantly hypermethylated genes are already repressed in pre-cancerous tissue and their hypermethylation does not directly contribute to cancer development via silencing.
