CXCR2 Inhibition Profoundly Suppresses Metastases and Augments Immunotherapy in Pancreatic Ductal Adenocarcinoma
Colin W. Steele,Saadia A. Karim,Joshua D.G. Leach,Peter Bailey,Rosanna Upstill-Goddard,Loveena Rishi,Mona Foth,Sheila Bryson,Karen McDaid,Zena Wilson,Catherine Anne Eberlein,Juliana Candido,Mairi Clarke,Colin Nixon,John T. Connelly,Nigel B. Jamieson,C. Ross Carter,Frances R. Balkwill,David K. Chang,T.R. Jeffry Evans,Douglas Strathdee,Andrew V. Biankin,Robert J. B. Nibbs,Simon T. Barry,Owen J. Sansom,Jennifer P. Morton +25 more
TL;DR: It is shown that CXCR2 signaling in the myeloid compartment can promote pancreatic tumorigenesis and is required for pancreatic cancer metastasis, making it an excellent therapeutic target.
Journal ArticleDOI
Inhibition of CXCR2 profoundly suppresses inflammation-driven and spontaneous tumorigenesis
Thomas Jamieson,Mairi Clarke,Colin W. Steele,Michael S. Samuel,Jens Neumann,Andreas Jung,David J. Huels,Michael F. Olson,Sudipto Das,Robert J. B. Nibbs,Owen J. Sansom +10 more
TL;DR: CXCR2 is a potent protumorigenic chemokine receptor that directs recruitment of tumor-promoting leukocytes into tissues during tumor-inducing and tumor-driven inflammation and may have therapeutic and prophylactic potential in the treatment of cancer.
Journal ArticleDOI
Epithelial NOTCH Signaling Rewires the Tumor Microenvironment of Colorectal Cancer to Drive Poor-Prognosis Subtypes and Metastasis.
Rene Jackstadt,Sander R. van Hooff,Joshua D.G. Leach,Xabier Cortes-Lavaud,Jeroen O. Lohuis,Rachel A. Ridgway,Valérie M. Wouters,Jatin Roper,Timothy J. Kendall,Campbell S.D. Roxburgh,Paul G. Horgan,Colin Nixon,Craig Nourse,Matthias Gunzer,William H. Clark,Ann Hedley,Ömer H. Yilmaz,Ömer H. Yilmaz,Mamunur Rashid,Peter Bailey,Andrew V. Biankin,Andrew D. Campbell,David J. Adams,Simon T. Barry,Colin W. Steele,Jan Paul Medema,Owen J. Sansom +26 more
TL;DR: Transcriptional profiling reveals that epithelial NOTCH1 signaling creates a tumor microenvironment reminiscent of poorly prognostic human CRC subtypes and drives metastasis through transforming growth factor (TGF) β-dependent neutrophil recruitment.
Journal ArticleDOI
Targeting the LOX/hypoxia axis reverses many of the features that make pancreatic cancer deadly: inhibition of LOX abrogates metastasis and enhances drug efficacy.
Bryan W. Miller,Jennifer P. Morton,Mark Pinese,Grazia Saturno,Nigel B. Jamieson,Ewan J. McGhee,Paul Timpson,Joshua D.G. Leach,Lynn McGarry,Emma Shanks,Peter Bailey,David K. Chang,Karin A. Oien,Saadia A. Karim,Amy Au,Colin W. Steele,C.R. Carter,Colin J. McKay,Kurt I. Anderson,Thomas R. Jeffry Evans,Richard Marais,Caroline J. Springer,Andrew V. Biankin,Janine T. Erler,Owen J. Sansom +24 more
TL;DR: It is found that LOX expression is upregulated in metastatic tumors from Pdx1‐Cre KrasG12D/+ Trp53R172H/+ (KPC) mice and that inhibition of LOX in these mice suppressed metastasis, and LOX inhibition is able to reverse many of the features that make PDAC inherently refractory to conventional therapies.
Journal ArticleDOI
Meta-analysis of oral antibiotics, in combination with preoperative intravenous antibiotics and mechanical bowel preparation the day before surgery, compared with intravenous antibiotics and mechanical bowel preparation alone to reduce surgical-site infections in elective colorectal surgery
TL;DR: The present systematic review and meta‐analysis aimed to investigate the effect of preoperative oral antibiotics and mechanical bowel preparation on SSI rates.
