Structure-based design of prefusion-stabilized SARS-CoV-2 spikes
Ching-Lin Hsieh,Jory A. Goldsmith,Jeffrey M. Schaub,Andrea M. DiVenere,Hung-Che Kuo,Kamyab Javanmardi,Kevin C. Le,Daniel Wrapp,Alison Gene-Wei Lee,Yutong Liu,Chia Wei Chou,Patrick O. Byrne,Christy K. Hjorth,Nicole V. Johnson,John Ludes-Meyers,Annalee W. Nguyen,Juyeon Park,Nianshuang Wang,Dzifa Amengor,Jason J. Lavinder,Gregory C. Ippolito,Jennifer A. Maynard,Ilya J. Finkelstein,Jason S. McLellan +23 more
TL;DR: High-yield production of a stabilized prefusion spike protein will accelerate the development of vaccines and serological diagnostics for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2).

Structure-based Design of Prefusion-stabilized SARS-CoV-2 Spikes
Ching-Lin Hsieh,Jory A. Goldsmith,Jeffrey M. Schaub,Andrea M. DiVenere,Hung-Che Kuo,Kamyab Javanmardi,Kevin C. Le,Daniel Wrapp,Alison Gene-Wei Lee,Yutong Liu,Chia-Wei Chou,Patrick O. Byrne,Christy K. Hjorth,Nicole V. Johnson,John Ludes-Meyers,Annalee W. Nguyen,Juyeon Park,Nianshuang Wang,Dzifa Amengor,Jennifer A. Maynard,Ilya J. Finkelstein,Jason S. McLellan +21 more
TL;DR: Over 100 structure-guided spike variants based upon a previously determined cryo-EM structure of the prefusion SARS-CoV-2 spike are designed and expressed, with the best variant, HexaPro, having six beneficial proline substitutions leading to ∼10-fold higher expression than its parental construct.

Identification of a conserved neutralizing epitope present on spike proteins from all highly pathogenic coronaviruses
Yuxin Huang,Annalee W. Nguyen,Ching-Lin Hsieh,Renato Menezes Silva,Olaluwoye Os,Wilen Re,Tamer S. Kaoud,Azouz Lr,Ahlam Qerqez,Kevin C. Le,Bohanon Al,Andrea M. DiVenere,Ying Liu,Alison Gene-Wei Lee,Dzifa Amengor,Dalby Kn,D’Arcy S,Jason S. McLellan,Jennifer A. Maynard +18 more
TL;DR: The authors identified residues 980-1006 in the flexible hinge region at the S2 apex as the 3A3 epitope, consistent with binding to natural and engineered spike variants, which may help guide design of pan-protective spike immunogens.
Journal ArticleDOI
Antibodies binding diverse pertactin epitopes protect mice from Bordetella pertussis infection
TL;DR: In this article , recombinant antibodies were found to protect multiple pertactin epitopes against Bordetella pertussis infection and significantly reduced lung colonization levels after sub-lethal challenge, with no antibody mediating antibodydependent complement-induced lysis.
Journal ArticleDOI
Identification of a conserved S2 epitope present on spike proteins from all highly pathogenic coronaviruses
Rui P. Silva,Yimin Huang,Annalee W. Nguyen,Ching-Lin Hsieh,Oladimeji Sunday Olaluwoye,Tamer S. Kaoud,Rebecca Wilen,Ahlam Qerqez,Jun-Gyu Park,Ahmed Magdy Khalil,Laura R. Azouz,Kevin C. Le,Amanda L Bohanon,Andrea M. DiVenere,Yutong Liu,Alison Gene-Wei Lee,Dzifa Amengor,Sophie R. Shoemaker,Shawn M. Costello,Eduardo A. Padlan,Susan Marqusee,Luis Martinez-Sobrido,Kevin N. Dalby,Sheena D'Arcy,Jason S. McLellan,Jennifer A. Maynard +25 more
TL;DR: In this paper , a conformational S2 domain epitope present only in the prefusion core of β-coronavirus was described, and a third class of S2 antibody was defined.
