The Putative Drp1 Inhibitor mdivi-1 Is a Reversible Mitochondrial Complex I Inhibitor that Modulates Reactive Oxygen Species
Evan A. Bordt,Pascaline Clerc,Brian A. Roelofs,Andrew J. Saladino,Andrew J. Saladino,Laszlo Tretter,Vera Adam-Vizi,Edward Cherok,Ahmed Khalil,Ahmed Khalil,Nagendra Yadava,Nagendra Yadava,Nagendra Yadava,Shealinna X. Ge,T. Chase Francis,Nolan W. Kennedy,Lora K. Picton,Tanya Kumar,Sruti Uppuluri,Alexandrea M. Miller,Kie Itoh,Mariusz Karbowski,Hiromi Sesaki,R. Blake Hill,Brian M. Polster +24 more
TL;DR: It is shown that mdivi-1 reversibly inhibits mitochondrial complex I-dependent O2 consumption and reverse electron transfer-mediated reactive oxygen species (ROS) production at concentrations (e.g., 50 μM) used to target mitochondrial fission.
Journal ArticleDOI
Mitochondrial structure, function and dynamics are temporally controlled by c-Myc.
J. Anthony Graves,Yudong Wang,Sunder Sims-Lucas,Edward Cherok,Kristi Rothermund,Maria F. Branca,Jennifer D. Elster,Donna Beer-Stolz,Bennett Van Houten,Jerry Vockley,Jerry Vockley,Edward V. Prochownik,Edward V. Prochownik +12 more
TL;DR: It is shown that re-expression of Myc in myc−/− fibroblasts is accompanied by a gradual accumulation of mitochondrial biomass and by increases in membrane polarization and mitochondrial fusion, which supports the idea that Myc affects mitochondrial mass by influencing both of these processes, albeit favoring the latter.
Journal ArticleDOI
Mitochondrial E3 ubiquitin ligase MARCH5 controls mitochondrial fission and cell sensitivity to stress-induced apoptosis through regulation of MiD49 protein.
Shan Xu,Edward Cherok,Shweta Das,Sunan Li,Brian A. Roelofs,Shealinna X. Ge,Brian M. Polster,Liron Boyman,W. Jonathan Lederer,Chunxin Wang,Mariusz Karbowski +10 more
TL;DR: Through ubiquitin- and proteasome-dependent degradation of MiD49, MARCH5 acts as a negative regulator of mitochondrial fission and thereby initiates a mechanism that affords the cell protection from stress-induced apoptosis.
Journal ArticleDOI
Novel regulatory roles of Mff and Drp1 in E3 ubiquitin ligase MARCH5-dependent degradation of MiD49 and Mcl1 and control of mitochondrial dynamics.
Edward Cherok,Shan Xu,Sunan Li,Shweta Das,W. Alex Meltzer,Michal Zalzman,Chunxin Wang,Mariusz Karbowski +7 more
TL;DR: In addition to their direct roles in mitochondrial fission, Mff and Drp1 act as regulatory factors and control mitochondria fission and fusion through a Ub-dependent mechanism.
Journal ArticleDOI
N-Myc overexpression increases cisplatin resistance in neuroblastoma via deregulation of mitochondrial dynamics.
Gabriella P. Casinelli,Jeff LaRosa,Manika Sharma,Edward Cherok,Swati Banerjee,Maria F. Branca,Lia R. Edmunds,Yudong Wang,Sunder Sims-Lucas,Luke Churley,Samantha Kelly,Ming Sun,Donna B. Stolz,J. Anthony Graves +13 more
TL;DR: It is found that N-Myc overexpressing cells are resistant to programmed cell death in response to exposure to low doses of cisplatin, and demonstrated that this was dependent on increased mitochondrial fusion.
