Journal ArticleDOI
Microglia development follows a stepwise program to regulate brain homeostasis
Orit Matcovitch-Natan,Deborah R. Winter,Amir Giladi,Stephanie Vargas Aguilar,Amit Spinrad,Sandrine Sarrazin,Sandrine Sarrazin,Sandrine Sarrazin,Hila Ben-Yehuda,Eyal David,Fabiola Zelada González,Fabiola Zelada González,Fabiola Zelada González,Pierre Perrin,Pierre Perrin,Pierre Perrin,Hadas Keren-Shaul,Meital Gury,David Lara-Astaiso,Christoph A. Thaiss,Merav Cohen,Keren Bahar Halpern,Kuti Baruch,Aleksandra Deczkowska,Erika Lorenzo-Vivas,Shalev Itzkovitz,Eran Elinav,Michael H. Sieweke,Michal Schwartz,Ido Amit +29 more
TL;DR: It is found that microglia from germ-free mice exhibited dysregulation of dozens of genes associated with the adult phase and immune response, including MAFB, which led to disruption of homeostasis in adulthood and increased expression of interferon and inflammation pathways.
Journal ArticleDOI
Chromatin state dynamics during blood formation
David Lara-Astiaso,Assaf Weiner,Erika Lorenzo-Vivas,Irina Zaretsky,Diego Jaitin,Eyal David,Hadas Keren-Shaul,Alexander Mildner,Deborah R. Winter,Steffen Jung,Nir Friedman,Ido Amit +11 more
TL;DR: A high-sensitivity indexing-first chromatin immunoprecipitation approach to profile the dynamics of four chromatin modifications across 16 stages of hematopoietic differentiation and reveals that lineage commitment involves de novo establishment of 17,035 lineage-specific enhancers.
Journal ArticleDOI
The Spectrum and Regulatory Landscape of Intestinal Innate Lymphoid Cells Are Shaped by the Microbiome
Meital Gury-BenAri,Christoph A. Thaiss,Nicolas Serafini,Nicolas Serafini,Deborah R. Winter,Amir Giladi,David Lara-Astiaso,Maayan Levy,Tomer-Meir Salame,Assaf Weiner,Eyal David,Hagit Shapiro,Mally Dori-Bachash,Meirav Pevsner-Fischer,Erika Lorenzo-Vivas,Hadas Keren-Shaul,Franziska Paul,Alon Harmelin,Gérard Eberl,Gérard Eberl,Shalev Itzkovitz,Amos Tanay,James P. Di Santo,James P. Di Santo,Eran Elinav,Ido Amit +25 more
TL;DR: The spectrum of transcriptional identities of small intestinal ILCs is characterized and how I LCs differentially integrate signals from the microbial microenvironment to generate phenotypic and functional plasticity is described.
Journal ArticleDOI
Genomic characterization of murine monocytes reveals C/EBPβ transcription factor dependence of Ly6C(-) cells
Alexander Mildner,Alexander Mildner,Jörg Schönheit,Amir Giladi,Eyal David,David Lara-Astiaso,Erika Lorenzo-Vivas,Franziska Paul,Louise Chappell-Maor,Josef Priller,Achim Leutz,Ido Amit,Steffen Jung +12 more
TL;DR: It is shown that monocyte differentiation occurred through de novo enhancer establishment and activation of pre‐established (poised) enhancers, and it is revealed that while Ly6C+ and Ly 6C− monocytes are homogeneous in steady state,Ly6Cint cells display heterogeneity.
Journal ArticleDOI
Subclinical atherosclerosis and accelerated epigenetic age mediated by inflammation: a multi-omics study.
Fátima Sánchez-Cabo,Valentin Fuster,Juan Carlos Silla-Castro,Erika Lorenzo-Vivas,Rebeca Álvarez,Sergio Callejas,Alberto Benguria,Estefanía Núñez,Belén Oliva,José M. Mendiguren,Marta Cortes-Canteli,Héctor Bueno,Vicente Andrés,José Marìa Ordovàs,Leticia Fernández-Friera,Antonio Jesus Quesada,Xavier Rossello,Jesús Vázquez,Ana Dopazo,Antonio Fernández-Ortiz,Borja Ibanez,José J. Fuster,Enrique Lara-Pezzi +22 more
TL;DR: In this article , the authors assessed the association of subclinical atherosclerosis with accelerated epigenetic age and investigated the underlying mechanisms mediating this association, concluding that systemic inflammation plays a key role in this association and reinforcing the relevance of interventions on inflammation to prevent cardiovascular disease.
