Journal ArticleDOI
Mycobacterial toxin induces analgesia in buruli ulcer by targeting the angiotensin pathways.
Estelle Marion,Estelle Marion,Ok-Ryul Song,Ok-Ryul Song,Ok-Ryul Song,Thierry Christophe,Jérémie Babonneau,Denis Philippe Cedric Fenistein,Joël Eyer,Frank Letournel,Daniel Henrion,Nicolas Clere,Vincent Paillé,Nathalie C. Guérineau,Jean-Paul Saint André,Philipp Gersbach,Karl-Heinz Altmann,Timothy P. Stinear,Yannick Comoglio,Guillaume Sandoz,Laurence Preisser,Yves Delneste,Edouard Yeramian,Laurent Marsollier,Laurent Marsollier,Priscille Brodin,Priscille Brodin +26 more
TL;DR: It is demonstrated here that M. ulcerans-induced hypoesthesia is instead achieved through a specific neurological pathway triggered by the secreted mycobacterial polyketide mycolactone, showing that my colactone elicits signaling through type 2 angiotensin II receptors (AT2Rs), leading to potassium-dependent hyperpolarization of neurons.
Journal ArticleDOI
Seasonal and Regional Dynamics of M. ulcerans Transmission in Environmental Context: Deciphering the Role of Water Bugs as Hosts and Vectors
Estelle Marion,Sara Eyangoh,Edouard Yeramian,Julien Marie C. Doannio,Jordi Landier,Jacques Aubry,Arnaud Fontanet,Christophe Rogier,Viviane Cassisa,Jane Cottin,Agnès Marot,Matthieu Eveillard,Yannick Kamdem,Pierre Legras,Caroline Deshayes,Jean-Paul Saint-André,Laurent Marsollier +16 more
TL;DR: This study demonstrated the presence of M. ulcerans in insect saliva, and provided a full picture of the ecology of transmission in terms of biodiversity and detailed specification of seasonal and regional dynamics, with large temporal heterogeneity in the insect tissue colonization rate.
Journal ArticleDOI
Geographic expansion of Buruli ulcer disease, Cameroon.
Estelle Marion,Jordi Landier,Pascal Boisier,Laurent Marsollier,Arnaud Fontanet,Philippe Le Gall,Jacques Aubry,Noumen Djeunga,Alphonse Umboock,Sara Eyangoh +9 more
TL;DR: The decreasing median age of patients since 2007 might be consistent with emergence of BU as a new disease in Bankim, and the possible role of environmental changes (building of a dam several years ago) in the expansion of BU in this area is hinted at.
Journal ArticleDOI
Kinetic analysis of Cas12a and Cas13a RNA-Guided nucleases for development of improved CRISPR-Based diagnostics
Eric Nalefski,Nidhi Patel,Philip J.Y. Leung,Zeba Islam,Remy M. Kooistra,Ishira Parikh,Estelle Marion,Gavin J. Knott,Gavin J. Knott,Jennifer A. Doudna,Anne-Laure M. Le Ny,Damian Madan +11 more
TL;DR: In this article, the authors examined the kinetics of single-turnover target and multi-turnoff trans-nuclease activities of both enzymes and showed that these enzymes are kinetically adapted to play distinct roles in bacterial adaptive immunity and show how kinetic analysis can be applied to CRISPR-based diagnostics.
Journal ArticleDOI
Mycolactone gene expression is controlled by strong SigA-like promoters with utility in studies of Mycobacterium ulcerans and buruli ulcer.
Nicholas J. Tobias,Torsten Seemann,Sacha J. Pidot,Jessica L. Porter,Laurent Marsollier,Estelle Marion,Franck Letournel,Tasnim Saifudin Zakir,Joseph Azuolas,John R. Wallace,Hui Hong,John K. Davies,Benjamin P Howden,Paul Dr Johnson,Grant A. Jenkin,Timothy P. Stinear +15 more
TL;DR: This is the first report to identify M. ulcerans toxin gene promoters, and the findings are used to develop M. Ulcerans-GFP, a strain in which fluorescence and toxin gene expression are linked, thus providing a tool for studying Buruli ulcer pathogenesis and potential transmission to humans.
