Journal ArticleDOI
HapX-mediated adaption to iron starvation is crucial for virulence of Aspergillus fumigatus.
Markus Schrettl,Nicola Beckmann,John J. Varga,Thorsten Heinekamp,Ilse D. Jacobsen,Christoph Jöchl,Tarek A. A. Moussa,Shaohua Wang,Fabio Gsaller,Michael Blatzer,Ernst R. Werner,William C. Niermann,Axel A. Brakhage,Hubertus Haas +13 more
TL;DR: In this paper, the bZIP-type regulator HapX is indispensable for the transcriptional remodeling required for adaption to iron starvation in the opportunistic fungal pathogen Aspergillus fumigatus.
Journal ArticleDOI
The Janus transcription factor HapX controls fungal adaptation to both iron starvation and iron excess
Fabio Gsaller,Peter Hortschansky,Sarah R. Beattie,Veronika Klammer,Katja Tuppatsch,Katja Tuppatsch,Beatrix E. Lechner,Nicole Rietzschel,Ernst R. Werner,Aaron A Vogan,Dawoon Chung,Ulrich Mühlenhoff,Masashi Kato,Robert A. Cramer,Axel A. Brakhage,Axel A. Brakhage,Hubertus Haas +16 more
TL;DR: This study uncovers a novel regulatory mechanism mediating both iron resistance and adaptation to iron starvation by the same transcription factor complex with activating and repressing functions depending on ambient iron availability.
Journal ArticleDOI
The negative cofactor 2 complex is a key regulator of drug resistance in Aspergillus fumigatus
Takanori Furukawa,Takanori Furukawa,Norman van Rhijn,Norman van Rhijn,Marcin G. Fraczek,Fabio Gsaller,Emma Davies,Paul Carr,Sara Gago,Sara Gago,Rachael Fortune-Grant,Rachael Fortune-Grant,Sayema Rahman,Sayema Rahman,Jane Mabey Gilsenan,Emma L Houlder,Caitlin H. Kowalski,Shriya Raj,Sanjoy Paul,Peter C. Cook,Josie E. Parker,Steve L. Kelly,Robert A. Cramer,Jean-Paul Latgé,Scott Moye-Rowley,Elaine Bignell,Elaine Bignell,Paul Bowyer,Paul Bowyer,Michael Bromley,Michael Bromley +30 more
TL;DR: A knockout library for 484 transcription factors in Aspergillus fumigatus is generated, and it is shown that loss of the NCT complex leads to cross-resistance to all primary and some salvage therapeutics without affecting pathogenicity.
Journal ArticleDOI
Sterol Biosynthesis and Azole Tolerance Is Governed by the Opposing Actions of SrbA and the CCAAT Binding Complex
Fabio Gsaller,Peter Hortschansky,Takanori Furukawa,Paul Carr,Bharat M. Rash,Javier Capilla,Christoph Müller,Franz Bracher,Paul Bowyer,Hubertus Haas,Axel A. Brakhage,Axel A. Brakhage,Michael Bromley +12 more
TL;DR: It is demonstrated that strains lacking a functional CBC are severely attenuated for pathogenicity in a pulmonary and systemic model of aspergillosis and inactivation of the CBC increased tolerance to different classes of drugs targeting ergosterol biosynthesis.
Journal ArticleDOI
The interplay between vacuolar and siderophore-mediated iron storage in Aspergillus fumigatus
Fabio Gsaller,Martin Eisendle,Beatrix E. Lechner,Markus Schrettl,Herbert Lindner,Daniela Müller,Stephan Geley,Hubertus Haas +7 more
TL;DR: It is shown that CccA-deficiency decreases iron resistance in particular in combination with derepressed iron uptake, while overproduction of CCCA increases iron resistance, which indicates that vacuolar rather than FC-mediated iron storage is the major iron detoxifying mechanism.
