Correction of ADA-SCID by Stem Cell Gene Therapy Combined with Nonmyeloablative Conditioning
Alessandro Aiuti,Shimon Slavin,Memet Aker,Francesca Ficara,Sara Deola,Alessandra Mortellaro,Shoshana Morecki,Grazia Andolfi,Antonella Tabucchi,Filippo Carlucci,Enrico Marinello,Federica Cattaneo,Sergio Vai,Paolo Servida,Roberto Miniero,Maria Grazia Roncarolo,Claudio Bordignon +16 more
TL;DR: Sustained engraftment of engineered HSCs with differentiation into multiple lineages resulted in increased lymphocyte counts, improved immune functions, and lower toxic metabolites, indicating the safety and efficacy of HSC gene therapy combined with nonmyeloablative conditioning for the treatment of SCID.
Journal ArticleDOI
Gene Therapy for Immunodeficiency Due to Adenosine Deaminase Deficiency
Alessandro Aiuti,Federica Cattaneo,Stefania Galimberti,Ulrike Benninghoff,Barbara Cassani,Luciano Callegaro,Samantha Scaramuzza,Grazia Andolfi,Massimiliano Mirolo,Immacolata Brigida,Antonella Tabucchi,Filippo Carlucci,Martha M. Eibl,Memet Aker,Shimon Slavin,Hamoud Al-Mousa,Abdulaziz Al Ghonaium,Alina Ferster,Andrea Duppenthaler,Luigi D. Notarangelo,Uwe Wintergerst,Rebecca H. Buckley,Marco Bregni,Sarah Marktel,Maria Grazia Valsecchi,Pier Luca Rossi,Fabio Ciceri,Roberto Miniero,Claudio Bordignon,Claudio Bordignon,Maria Grazia Roncarolo +30 more
TL;DR: Gene therapy, combined with reduced-intensity conditioning, is a safe and effective treatment for SCID in patients with ADA deficiency and effective protection against infections and improvement in physical development made a normal lifestyle possible.
Journal ArticleDOI
Immune reconstitution in ADA-SCID after PBL gene therapy and discontinuation of enzyme replacement.
Alessandro Aiuti,Sergio Vai,Alessandra Mortellaro,Giulia Casorati,Francesca Ficara,Grazia Andolfi,Giuliana Ferrari,Antonella Tabucchi,Filippo Carlucci,Hans D. Ochs,Luigi D. Notarangelo,Maria Grazia Roncarolo,Claudio Bordignon +12 more
TL;DR: Immune reconstitution in ADA-SCID after PBL gene therapy and discontinuation of enzyme replacement is shown to occur after two weeks after treatment with PBL.
Journal ArticleDOI
Update on the safety and efficacy of retroviral gene therapy for immunodeficiency due to adenosine deaminase deficiency
Maria Pia Cicalese,Francesca Ferrua,Laura Castagnaro,Roberta Pajno,Federica Barzaghi,Stefania Giannelli,Francesca Dionisio,Immacolata Brigida,Marco Bonopane,Miriam Casiraghi,Antonella Tabucchi,Filippo Carlucci,Eyal Grunebaum,Mehdi Adeli,Robbert G. M. Bredius,Jennifer M. Puck,Polina Stepensky,Ilhan Tezcan,Katie Rolfe,Erika H. De Boever,Rickey R. Reinhardt,Jonathan Appleby,Fabio Ciceri,Maria Grazia Roncarolo,Alessandro Aiuti +24 more
TL;DR: This investigation investigated the medium-term outcome of gene therapy (GT) in 18 patients with ADA-SCID for whom an HLA-identical family donor was not available; most were not responding well to ERT.
Journal ArticleDOI
Subnormothermic machine perfusion protects steatotic livers against preservation injury: A potential for donor pool increase?
Mariapia Vairetti,Andrea Ferrigno,Filippo Carlucci,Antonella Tabucchi,Vittoria Rizzo,Eleonora Boncompagni,Daniele Neri,Enrico Gringeri,Isabel Freitas,Umberto Cillo +9 more
TL;DR: MP at 20°C improves cell survival and leads to a marked improvement in hepatic preservation of steatotic livers as compared with cold storage, substantiated by better morphology, higher glycogen content, and lower reactive oxygen species production by sinusoidal cells in Steatotic liver submitted to MP at20°C versus cold storage.
