Transfusion independence and HMGA2 activation after gene therapy of human β-thalassaemia
Marina Cavazzana-Calvo,Emmanuel Payen,Olivier Negre,Gary P. Wang,Kathleen M. Hehir,Floriane Fusil,Julian D. Down,Maria Denaro,Troy Brady,Karen A. Westerman,Resy Cavallesco,Beatrix Gillet-Legrand,Laure Caccavelli,Laure Caccavelli,Riccardo Sgarra,Leila Maouche-Chretien,Françoise Bernaudin,Robert Girot,Ronald Dorazio,Geert Jan Mulder,Axel Polack,Arthur Bank,Jean Soulier,Jérôme Larghero,Nabil Kabbara,Bruno Dalle,Bernard Gourmel,Gérard Socié,Stany Chrétien,Stany Chrétien,Nathalie Cartier,Patrick Aubourg,Alain Fischer,Alain Fischer,Kenneth Cornetta,Frédéric Galactéros,Yves Beuzard,Eliane Gluckman,Frederick D. Bushman,Salima Hacein-Bey-Abina,Salima Hacein-Bey-Abina,Philippe Leboulch +41 more
TL;DR: It is shown that, 33 months after lentiviral β-globin gene transfer, an adult patient with severe βE/β0-thalassaemia dependent on monthly transfusions since early childhood has become transfusion independent for the past 21 months.
Journal ArticleDOI
Impairment of mycobacterial immunity in human interleukin-12 receptor deficiency
Frédéric Altare,Anne Durandy,David A. Lammas,J.F. Emile,S Lamhamedi,Françoise Le Deist,Pam Drysdale,Emmanuelle Jouanguy,Rainer Doffinger,Françoise Bernaudin,Olle Jeppsson,Jared Gollob,Edgar Meinl,Antony W. Segal,Alain Fischer,Dinakantha S. Kumararatne,Jean-Laurent Casanova +16 more
TL;DR: Interleukin-12 (IL-12) receptor deficiency was found in otherwise healthy individuals with mycobacterial infections, and mature granulomas were seen, surrounded by T cells and centered with epithelioid and multinucleated giant cells, yet reduced IFN-gamma concentrations were found to be secreted by activated natural killer and T cells.
Journal ArticleDOI
Bone marrow transplantation for sickle cell disease.
Mark C. Walters,M. Patience,Wendy M. Leisenring,Eckman,Scott Jp,William C. Mentzer,Sally C. Davies,Kwaku Ohene-Frempong,Françoise Bernaudin,Dana C. Matthews,R Storb,Keith M. Sullivan +11 more
TL;DR: Allogeneic stem-cell transplantation can be curative in young patients with symptomatic sickle cell disease and Kaplan-Meier estimates of survival and event-free survival at four years were 91 percent and 73 percent, respectively.
Journal ArticleDOI
Long-term results of related myeloablative stem-cell transplantation to cure sickle cell disease
Françoise Bernaudin,Gérard Socié,Mathieu Kuentz,Sylvie Chevret,Michel Duval,Yves Bertrand,Jean-Pierre Vannier,Karima Yakouben,Isabelle Thuret,Pierre Bordigoni,Alain Fischer,Patrick Lutz,Jean-Louis Stephan,Nathalie Dhedin,Emmanuel Plouvier,Geneviève Margueritte,Dominique Bories,Suzanne Verlhac,Helene Esperou,Lena Coic,Jean-Paul Vernant,Eliane Gluckman +21 more
TL;DR: Results indicate that HLA-identical sibling HSCT after myeloablative conditioning with ATG should be considered as a standard of care for SCD children who are at high risk for stroke.
Journal ArticleDOI
Impact of bone marrow transplantation for symptomatic sickle cell disease: an interim report
Mark C. Walters,Rainer Storb,Melinda Patience,Wendy M. Leisenring,Terri Taylor,Jean E. Sanders,George E. Buchanan,Zora R. Rogers,Patricia A. Dinndorf,Sally C. Davies,Irene Roberts,Rosarita Dickerhoff,Andrew M. Yeager,Hsu Lewis,Joanne Kurtzberg,Kwaku Ohene-Frempong,Nancy Bunin,Françoise Bernaudin,Wing Yen Wong,J. Paul Scott,David A. Margolis,Elliott Vichinsky,Donna A. Wall,Allen S. Wayne,Charles H. Pegelow,Rupa Redding-Lallinger,Joseph Wiley,Martin R. Klemperer,William C. Mentzer,Franklin O. Smith,Keith M. Sullivan +30 more
TL;DR: Data confirm that allogenic bone marrow transplantation establishes normal erythropoiesis and is associated with improved growth and stable CNS imaging and pulmonary function in most patients.
