Genomic maps of long noncoding RNA occupancy reveal principles of RNA-chromatin interactions.
TL;DR: ChIRP-seq of three lncRNAs reveal that RNA occupancy sites in the genome are focal, sequence-specific, and numerous, and generally applicable to illuminate the intersection of RNA and chromatin with newfound precision genome wide.
Journal ArticleDOI
Physical plasticity of the nucleus in stem cell differentiation
TL;DR: Micromanipulation methods show that nuclei in human embryonic stem cells are highly deformable and stiffen 6-fold through terminal differentiation, and that nucleo-skeletal components in human adult stem cells possess an intermediate stiffness and deform irreversibly.
Journal ArticleDOI
T helper type 1 and 17 cells determine efficacy of interferon-beta in multiple sclerosis and experimental encephalomyelitis.
Robert C. Axtell,Brigit A. de Jong,Brigit A. de Jong,Brigit A. de Jong,Katia Boniface,Laura F van der Voort,Roopa Bhat,Patrizia De Sarno,Rodrigo Naves,May H. Han,Franklin L. Zhong,Jim G Castellanos,Robert D. Mair,Athena Christakos,Ilan Kolkowitz,Liat Katz,Joep Killestein,Chris H. Polman,Rene de Waal Malefyt,Lawrence Steinman,Chander Raman +20 more
TL;DR: Congruence in outcome between responses to IFN-β in experimental autoimmune encephalomyelitis (EAE) and relapsing-remitting multiple sclerosis (RRMS) is found and a high IL-17F concentration in the serum of people with RRMS is associated with nonresponsiveness to therapy with IFn-β.
Journal ArticleDOI
Germline NLRP1 Mutations Cause Skin Inflammatory and Cancer Susceptibility Syndromes via Inflammasome Activation

Robinson,Ildikó Szeverényi,Takuya Takeichi,Takuya Takeichi,Reshmaa Balaji,Aristotle Lau,Hazel Tye,Hazel Tye,Keya Roy,Carine Bonnard,Patricia J. Ahl,L. A. Jones,Paul J. Baker,Paul J. Masters,Vincent Soler,Salma Samir Omar,John A. McGrath,Roxana Ioana Nedelcu,Moez Gribaa,Mohamed Denguezli,Ali Saad,Sebastian Hiller,Bruno Reversade +38 more
TL;DR: It is found that germline mutations in the inflammasome sensor NLRP1 cause two overlapping skin disorders: multiple self-healing palmoplantar carcinoma (MSPC) and familial keratosis lichenoides chronica (FKLC) and the first genetic evidence linkingNLRP1 to skin inflammatory syndromes and skin cancer predisposition is provided.
Journal ArticleDOI
TPP1 OB-Fold Domain Controls Telomere Maintenance by Recruiting Telomerase to Chromosome Ends
Franklin L. Zhong,Luis F.Z. Batista,Adam Freund,Matthew F. Pech,Andrew S. Venteicher,Steven E. Artandi +5 more
TL;DR: It is shown that the OB-fold domain of the telomere-binding protein TPP1 recruits telomersase to telomeres through an association with the telomerase reverse transcriptase TERT, and this data defines a potential interface for telomerases-TPP1 interaction required forTelomere maintenance and implicate defective telomer enzyme recruitment in telomerasing-related disease.
