Journal ArticleDOI
Identification of SARS-CoV-2 inhibitors using lung and colonic organoids.
Yuling Han,Xiaohua Duan,Xiaohua Duan,Liuliu Yang,Benjamin E. Nilsson-Payant,Pengfei Wang,Fuyu Duan,Xuming Tang,Tomer M. Yaron,Tuo Zhang,Skyler Uhl,Yaron Bram,Chanel Richardson,Jiajun Zhu,Zeping Zhao,David Redmond,Sean Houghton,Duc-Huy T. Nguyen,Dong Xu,Xing Wang,Jose Jessurun,Alain C. Borczuk,Yaoxing Huang,Jared L. Johnson,Yuru Liu,Jenny Xiang,Hui Wang,Lewis C. Cantley,Benjamin R. tenOever,David D. Ho,Fong Cheng Pan,Todd Evans,Huanhuan Joyce Chen,Huanhuan Joyce Chen,Robert E. Schwartz,Shuibing Chen +35 more
TL;DR: This work developed a lung organoid model using human pluripotent stem cells (hPSC-LOs) and performed a high-throughput screen of drugs approved by the FDA and identified entry inhibitors of SARS-CoV-2, including imatinib, mycophenolic acid and quinacrine dihydrochloride.

Identification of Candidate COVID-19 Therapeutics using hPSC-derived Lung Organoids
Yuling Han,Liuliu Yang,Xiaohua Duan,Fuyu Duan,Benjamin E. Nilsson-Payant,Tomer M. Yaron,Pengfei Wang,Xuming Tang,Tuo Zhang,Zeping Zhao,Yaron Bram,David Redmond,Sean Houghton,Duc T. Nguyen,Dong Xu,Xing Wang,Skyler Uhl,Yaoxing Huang,Jared L. Johnson,Jenny Xiang,Hui Wang,Fong Cheng Pan,Lewis C. Cantley,Benjamin R. tenOever,David D. Ho,Todd Evans,Robert E. Schwartz,Huanhuan Joyce Chen,Shuibing Chen +28 more
TL;DR: It is demonstrated that hPSC-derived lung cells infected by SARS-CoV-2 can model human COVID-19 disease and provide a valuable resource to screen for FDA-approved drugs that might be repurposed and should be considered for CO VID-19 clinical trials.

Modeling COVID-19 with Human Pluripotent Stem Cell-Derived Cells Reveals Synergistic Effects of Anti-inflammatory Macrophages with ACE2 Inhibition Against SARS-CoV-2
Fuyu Duan,Liyan Guo,Liuliu Yang,Yuling Han,Abhimanyu Thakur,Benjamin E. Nilsson-Payant,Pengfei Wang,Zhao Zhang,Chui Yan Ma,Xiaoya Zhou,Teng Han,Tuo Zhang,Xing Wang,Dong Xu,Xiaohua Duan,Jenny Xiang,Hung-Fat Tse,Can Liao,Weiren Luo,Fang-Ping Huang,Ya-Wen Chen,Todd Evans,Robert E. Schwartz,Benjamin R. tenOever,David D. Ho,Shuibing Chen,Qizhou Lian,Huanhuan Joyce Chen +27 more
TL;DR: Direct differentiation of human pluripotent stem cells is used to establish a lung and macrophage co-culture system and model the host-pathogen interaction and immune response caused by SARS-CoV-2 infection, suggesting a potential therapeutic strategy by boosting anti-inflammatory action of macrophages at an early stage of infection.
Differential effects of macrophage subtypes on SARS-CoV-2 infection in a human pluripotent stem cell-derived model

Zhou,Shuo Han,Teng-Long Han,Wei Na Zhang,Adrian Y. Tan,Tuo Zhang,Xing Wang,Donghui Xu,Jenny Xiang,Aimin Xu,Can Liao,Fang-Ping Huang,Ya-Wen Chen,Jie Na,Glenn Randall,Hung-Fat Tse,Zhiwei Chen,Yin Chen,Huanhuan Joyce Chen +31 more
TL;DR: A lung and macrophage co-culture system derived from human pluripotent stem cells is established, modeling the host-pathogen interaction in SARS-CoV-2 infection in vitro, finding that both classically polarized macrophages (M1) and alternatively polarized macophage (M2) have inhibitory effects on Sars-CoVs infection.

FGF21–Sirtuin 3 Axis Confers the Protective Effects of Exercise Against Diabetic Cardiomyopathy by Governing Mitochondrial Integrity
Leigang Jin,Leiluo Geng,Lei Ying,Lei Shu,Kevin Ye,Ranyao Yang,Yan Liu,Yao Wang,Yin Cai,Xue Jiang,Qin Wang,Xingqun Yan,Boya Liao,Jie Liu,Fuyu Duan,Gary Sweeney,Connie W.H. Woo,Yu Wang,Zhengyuan Xia,Qizhou Lian,Aimin Xu +20 more
TL;DR: The FGF21-SIRT3 axis mediates the protective effects of exercise against DCM by preserving mitochondrial integrity and represents a potential therapeutic target for DCM.
