A Human Pluripotent Stem Cell-based Platform to Study SARS-CoV-2 Tropism and Model Virus Infection in Human Cells and Organoids.
Liuliu Yang,Yuling Han,Benjamin E. Nilsson-Payant,Vikas Gupta,Pengfei Wang,Xiaohua Duan,Xiaohua Duan,Xuming Tang,Jiajun Zhu,Zeping Zhao,Fabrice Jaffré,Tuo Zhang,Tae Wan Kim,Oliver Harschnitz,David Redmond,Sean Houghton,Chengyang Liu,Ali Naji,Gabriele Ciceri,Sudha R Guttikonda,Yaron Bram,Duc-Huy T. Nguyen,Michele Cioffi,Vasuretha Chandar,Daisy A. Hoagland,Yaoxing Huang,Jenny Xiang,Hui Wang,Hui Wang,David Lyden,Alain C. Borczuk,Huanhuan Joyce Chen,Lorenz Studer,Fong Cheng Pan,David D. Ho,Benjamin R. tenOever,Todd Evans,Robert E. Schwartz,Shuibing Chen +38 more
TL;DR: It is found that human pancreatic beta cells and liver organoids are highly permissive to SARS-CoV-2 infection, further validated using adult primary human islets and adult hepatocyte and cholangiocyte organoids.
Journal ArticleDOI
Identification of SARS-CoV-2 inhibitors using lung and colonic organoids.
Yuling Han,Xiaohua Duan,Xiaohua Duan,Liuliu Yang,Benjamin E. Nilsson-Payant,Pengfei Wang,Fuyu Duan,Xuming Tang,Tomer M. Yaron,Tuo Zhang,Skyler Uhl,Yaron Bram,Chanel Richardson,Jiajun Zhu,Zeping Zhao,David Redmond,Sean Houghton,Duc-Huy T. Nguyen,Dong Xu,Xing Wang,Jose Jessurun,Alain C. Borczuk,Yaoxing Huang,Jared L. Johnson,Yuru Liu,Jenny Xiang,Hui Wang,Lewis C. Cantley,Benjamin R. tenOever,David D. Ho,Fong Cheng Pan,Todd Evans,Huanhuan Joyce Chen,Huanhuan Joyce Chen,Robert E. Schwartz,Shuibing Chen +35 more
TL;DR: This work developed a lung organoid model using human pluripotent stem cells (hPSC-LOs) and performed a high-throughput screen of drugs approved by the FDA and identified entry inhibitors of SARS-CoV-2, including imatinib, mycophenolic acid and quinacrine dihydrochloride.
Posted ContentDOI
Identification of Candidate COVID-19 Therapeutics using hPSC-derived Lung Organoids
Yuling Han,Liuliu Yang,Xiaohua Duan,Fuyu Duan,Benjamin E. Nilsson-Payant,Tomer M. Yaron,Pengfei Wang,Xuming Tang,Tuo Zhang,Zeping Zhao,Yaron Bram,David Redmond,Sean Houghton,Duc T. Nguyen,Dong Xu,Xing Wang,Skyler Uhl,Yaoxing Huang,Jared L. Johnson,Jenny Xiang,Hui Wang,Fong Cheng Pan,Lewis C. Cantley,Benjamin R. tenOever,David D. Ho,Todd Evans,Robert E. Schwartz,Huanhuan Joyce Chen,Shuibing Chen +28 more
TL;DR: It is demonstrated that hPSC-derived lung cells infected by SARS-CoV-2 can model human COVID-19 disease and provide a valuable resource to screen for FDA-approved drugs that might be repurposed and should be considered for CO VID-19 clinical trials.
Posted ContentDOI
Combined small molecule treatment accelerates timing of maturation in human pluripotent stem cell-derived neurons
Emiliano Hergenreder,Ya. O. Zorina,Zeping Zhao,Hermany Munguba,Elizabeth L. Calder,Arianna Baggiolini,Andrew P. Minotti,Ryan M. Walsh,Conor Liston,Joshua Levitz,Ralph Garippa,Shuibing Chen,Gabriele Ciceri,Lorenz Studer +13 more
TL;DR: It is demonstrated that the maturation of multiple hPSC-derived cell types can be enhanced by simple pharmacological intervention and suggests that some of the mechanisms controlling the timing of human maturation are shared across lineages.
Journal ArticleDOI
A human iPSC-array-based GWAS identifies a virus susceptibility locus in the NDUFA4 gene and functional variants
Yuling Han,Lei Tan,Ting Zhou,Liuliu Yang,Lucia Carrau,Lauretta A. Lacko,Mohsan Saeed,Jiajun Zhu,Zeping Zhao,Benjamin E. Nilsson-Payant,Filipe Tenorio Lira Neto,Clare W Cahir,Alice Giani,Jin Chou Chai,Xue Dong,Dorota Moroziewicz,Daniel Paull,Tuo Zhang,So Yeon Koo,Christina Tan,Ron Danziger,Qian Ba,Lingling Feng,Zhengming Chen,Aaron Zhong,Gilbert J. Wise,Jenny Xiang,Hui Wang,Robert E. Schwartz,Benjamin R. tenOever,Scott Noggle,Charles M. Rice,Qibin Qi,Todd Evans,Shuibing Chen +34 more
TL;DR: In this paper , a human-induced pluripotent stem cell (hiPSC)-based screening strategy was used to link human genetics with viral infectivity, and a genome-wide association study (GWAS) identified a cluster of single-nucleotide polymorphisms (SNPs) in a cis-regulatory region of the NDUFA4 gene, which was associated with susceptibility to Zika virus infection.
