Journal ArticleDOI
Beta cells can be generated from endogenous progenitors in injured adult mouse pancreas.
Xiaobo Xu,Joke D'Hoker,Joke D'Hoker,Geert Stange,Geert Stange,Stefan Bonné,Stefan Bonné,Nico De Leu,Nico De Leu,Xiangwei Xiao,Xiangwei Xiao,Mark Van de Casteele,Mark Van de Casteele,Georg Mellitzer,Georg Mellitzer,Zhidong Ling,Zhidong Ling,Daniel Pipeleers,Daniel Pipeleers,Luc Bouwens,Luc Bouwens,Raphael Scharfmann,Raphael Scharfmann,Raphael Scharfmann,Gerard Gradwohl,Gerard Gradwohl,Gerard Gradwohl,Harry Heimberg,Harry Heimberg,Harry Heimberg +29 more
TL;DR: Guided by the expression of Neurogenin 3 (Ngn3), the earliest islet cell-specific transcription factor in embryonic development, it is shown that beta cell progenitors can be activated in injured adult mouse pancreas and are located in the ductal lining.
Journal ArticleDOI
Genetic determinants of pancreatic epsilon-cell development.
R S Heller,Marjorie Jenny,Patrick Collombat,Ahmed Mansouri,Catherine Tomasetto,Ole D. Madsen,Georg Mellitzer,Gérard Gradwohl,Palle Serup +8 more
TL;DR: It is demonstrated that the development of ghrelin-producing cells is differentially dependent on Neurogenin3 in different domains of the gastrointestinal tract and that, in the endocrine pancreas, epsilon-cell genesis does not require Arx or Pax4 activities but is antagonized by Pax6.
Journal ArticleDOI
IA1 is NGN3-dependent and essential for differentiation of the endocrine pancreas
Georg Mellitzer,Stefan Bonné,Reini F. Luco,Mark Van de Casteele,Nathalie Lenne-Samuel,Patrick Collombat,Ahmed Mansouri,Jacqueline E. Lee,Michael S. Lan,Daniel Pipeleers,Finn Cilius Nielsen,Jorge Ferrer,Gérard Gradwohl,Harry Heimberg +13 more
TL;DR: IA1 is a novel Ngn3‐regulated factor required for normal differentiation of pancreatic endocrine cells, and forms a novel branch in the Ngn2‐dependent endocrinogenic transcription factor network.
Journal ArticleDOI
Pancreatic Islet Progenitor Cells in Neurogenin 3-Yellow Fluorescent Protein Knock-Add-On Mice
Georg Mellitzer,Mercè Martín,Marjorie Sidhoum-Jenny,Christophe Orvain,Jochen Barths,Philip A. Seymour,Maike Sander,Gérard Gradwohl +7 more
TL;DR: Endocrine progenitors can form de novo and expand, in vitro, in the absence of signals from the surrounding mesenchyme, suggesting that endocrine commitment is a default pathway.
Journal ArticleDOI
Spatial and temporal 'knock down' of gene expression by electroporation of double-stranded RNA and morpholinos into early postimplantation mouse embryos.
TL;DR: It is demonstrated for the first time that dsRNA and morpholino technologies can be successfully applied in early postimplantation mouse embryos to specifically knock down gene expression.
