Journal ArticleDOI
Vorinostat plus tacrolimus and mycophenolate to prevent graft-versus-host disease after related-donor reduced-intensity conditioning allogeneic haemopoietic stem-cell transplantation: a phase 1/2 trial
Sung Won Choi,Thomas Braun,Lawrence Chang,James L.M. Ferrara,Attaphol Pawarode,John M. Magenau,Guoqing Hou,Jan H. Beumer,John E. Levine,S. A. Goldstein,Daniel R. Couriel,Keith Stockerl-Goldstein,Oleg I. Krijanovski,Carrie L. Kitko,Gregory A. Yanik,Michael H. Lehmann,Isao Tawara,Yaping Sun,Sophie Paczesny,Markus Y. Mapara,Charles A. Dinarello,John F. DiPersio,Pavan Reddy +22 more
TL;DR: Administration of vorinostat in combination with standard GVHD prophylaxis after related-donor reduced-intensity conditioning haemopoietic stem-cell transplantation is safe and is associated with a lower than expected incidence of severe acute GV HD.
Journal ArticleDOI
Histone deacetylase inhibition regulates inflammation and enhances Tregs after allogeneic hematopoietic cell transplantation in humans
Sung Won Choi,Erin Gatza,Guoqing Hou,Yaping Sun,Joel Whitfield,Yeohan Song,Katherine Oravecz-Wilson,Isao Tawara,Charles A. Dinarello,Pavan Reddy +9 more
TL;DR: It is demonstrated that HDAC inhibition reduces inflammatory responses of PBMC but enhances Tregs after allo-HCT, andHDAC inhibition also increased signal transducer and activator of transcription 3 acetylation and induced indoleamine-2,3-dioxygenase.
Journal ArticleDOI
Siglec-G–CD24 axis controls the severity of graft-versus-host disease in mice
Tomomi Toubai,Guoqing Hou,Nathan Mathewson,Chen Liu,Ying Wang,Katherine Oravecz-Wilson,Emily Cummings,Corinne Rossi,Rebecca Evers,Yaping Sun,Julia Wu,Sung Won Choi,Dexing Fang,Pan Zheng,Yang Liu,Pavan Reddy +15 more
TL;DR: It is reported that Siglec-G expression on host APCs, specifically on hematopoietic cells, negatively regulates GVHD in multiple clinically relevant murine models and suggested that enhancing this interaction may represent a novel strategy for mitigating GV HD.
Journal ArticleDOI
Neddylation plays an important role in the regulation of murine and human dendritic cell function
Nathan Mathewson,Tomomi Toubai,Steven Kapeles,Yaping Sun,Katherine Oravecz-Wilson,Hiroya Tamaki,Ying Wang,Guoqing Hou,Yi Sun,Pavan Reddy +9 more
TL;DR: Inhibition of neddylation suppressed the release of proinflammatory cytokines by DCs in response to Toll-like receptor, nucleotide oligomerization domain- like receptor, and noninfectious CD40L stimulation, and could be a novel strategy for mitigating immune-mediated disease processes.
Journal ArticleDOI
Targeting Histone Deacetylases As a New Strategy for Graft Versus Host Disease Prevention
Sung Won Choi,John F. DiPersio,Thomas Braun,Guoqing Hou,Keith Stockerl-Goldstein,Isao Tawara,John E. Levine,Yaping Sun,Daniel R. Couriel,Oleg I. Krijanovski,Lawrence Chang,John M. Magenau,Attaphol Pawarode,Carrie L. Kitko,Sophie Paczesny,Edward Peres,Shin Mineishi,Gregory A. Yanik,Michael H. Lehmann,Charles A. Dinarello,James L.M. Ferrara,Pavan Reddy +21 more
TL;DR: The hypothesis that HDAC inhibition will be safe and reduce the severity of GVHD in patients undergoing matched related donor (MRD) reduced intensity conditioning (RIC) allogeneic HSCT is tested in a first-in-human clinical trial.
