MYC Drives Progression of Small Cell Lung Cancer to a Variant Neuroendocrine Subtype with Vulnerability to Aurora Kinase Inhibition
Gurkan Mollaoglu,Matthew R. Guthrie,Stefanie Böhm,Johannes Brägelmann,Ismail Can,Paul M. Ballieu,Annika Marx,Julie George,Christine Heinen,Milind D. Chalishazar,Haixia Cheng,Abbie S. Ireland,Kendall E. Denning,Anandaroop Mukhopadhyay,Jeffery M. Vahrenkamp,Kristofer C. Berrett,Timothy L. Mosbruger,Jun Wang,Jessica L. Kohan,Mohamed E. Salama,Benjamin L. Witt,Martin Peifer,Roman K. Thomas,Roman K. Thomas,Jason Gertz,Jane E. Johnson,Adi F. Gazdar,Robert J. Wechsler-Reya,Martin L. Sos,Trudy G. Oliver +29 more
TL;DR: It is shown that Myc expression cooperates with Rb1 and Trp53 loss in the mouse lung to promote aggressive, highly metastatic tumors, that are initially sensitive to chemotherapy followed by relapse, similar to human SCLC.
Journal ArticleDOI
Recurrent WNT pathway alterations are frequent in relapsed small cell lung cancer
Alex H. Wagner,Siddhartha Devarakonda,Zachary L. Skidmore,Kilannin Krysiak,Avinash Ramu,Lee Trani,Jason Kunisaki,Ashiq Masood,Saiama N. Waqar,Nicholas C. Spies,Daniel Morgensztern,Jason Waligorski,Jennifer Ponce,Robert S. Fulton,Leonard B. Maggi,Jason D. Weber,Mark A. Watson,Christopher J. O’Conor,Jon H. Ritter,Rachelle R. Olsen,Haixia Cheng,Anandaroop Mukhopadhyay,Ismail Can,Melissa H. Cessna,Trudy G. Oliver,Elaine R. Mardis,Rick K. Wilson,Malachi Griffith,Obi L. Griffith,Ramaswamy Govindan +29 more
TL;DR: The authors analyse the genomic and transcriptomic landscape of primary and relapsed SCLC patients as well as in vitro models, and discover that activation of WNT signalling can drive chemotherapy resistance.
Journal ArticleDOI
The p300 Inhibitor A-485 Exerts Antitumor Activity in Growth Hormone Pituitary Adenoma
Chenxing Ji,Wen Xu,Hong Ding,Zhengyuan Chen,Chengzhang Shi,Jie Han,Liang Yu,Nidan Qiao,Yichao Zhang,Xiao-Tao Cao,Xiang Zhou,Haixia Cheng,Huijin Feng,Cheng Luo,Zhiyu Li,Bing-Bing Zhou,Zhao Ye,Yao Zhao +17 more
TL;DR: It is demonstrated that inhibition of HAT p300 by its selective inhibitor A-485 is a promising therapy for GHPA and could downregulate the expression or activity of several oncogenes, such as genes in the Pttg1, c-Myc, cAMP and PI3K/AKT/mTOR signaling pathways, which are crucial for PA tumorigenesis and progression.
Journal ArticleDOI
Single-cell transcriptome reveals cellular hierarchies and guides p-EMT-targeted trial in skull base chordoma
Qilin Zhang,Lijiang Fei,Rui Han,Ruofan Huang,Yongfei Wang,Hongyu Chen,Boyuan Yao,Nidan Qiao,Zhe Wang,Zengyi Ma,Zhao Ye,Yichao Zhang,Weiwei Wang,Ye Wang,Lingxia Kong,Xuefei Shou,Xiao-Tao Cao,Xiang Zhou,Ming Shen,Haixia Cheng,Zhenwei Yao,Chao Zhang,Guoji Guo,Yao Zhao +23 more
TL;DR: Wang et al. as mentioned in this paper identified a cluster of stem-like SBC cells that tended to be distributed in the inferior part of the tumor, and they further found that this cell cluster is marked by cathepsin L (CTSL ), a gene involved in the packaging of telomere ends, and may be responsible for radioresistance.
Journal ArticleDOI
Single-cell sequencing identifies differentiation-related markers for molecular classification and recurrence prediction of PitNET
Qilin Zhang,Boyuan Yao,Xin Long,Zhengyuan Chen,Min He,Yue Wu,Nidan Qiao,Zengyi Ma,Zhao Ye,Yichao Zhang,Shun Yao,Ye Wang,Haixia Cheng,Hong Chen,Hongying Ye,Yongfei Wang,Yimin Li,Jianhua Chen,Zhaoyun Zhang,Fang Guo,Yao Zhao +20 more
TL;DR: In this paper , a comparative analysis of single-cell RNA sequencing data from 3 anterior pituitary glands and 21 PitNETs was performed to comprehensively reveal the origin and differentiation status of PitNET.
