Journal ArticleDOI
eIF5A promotes translation of polyproline motifs.
Erik Gutierrez,Byung-Sik Shin,Christopher J. Woolstenhulme,Joo-Ran Kim,Preeti Saini,Allen R. Buskirk,Thomas E. Dever +6 more
TL;DR: EIF5A, like its bacterial ortholog EFP, is proposed to stimulate the peptidyl transferase activity of the ribosome and facilitate the reactivity of poor substrates like Pro.
Journal ArticleDOI
eIF2γ Mutation that Disrupts eIF2 Complex Integrity Links Intellectual Disability to Impaired Translation Initiation
Guntram Borck,Byung-Sik Shin,Barbara Stiller,Aviva Mimouni-Bloch,Holger Thiele,Joo-Ran Kim,Meghna Thakur,Cindy Skinner,Lara Aschenbach,Pola Smirin-Yosef,Adi Har-Zahav,Gudrun Nürnberg,Janine Altmüller,Peter Frommolt,Kay Hofmann,Osnat Konen,Peter Nürnberg,Arnold Munnich,Charles E. Schwartz,Doron Gothelf,Doron Gothelf,Laurence Colleaux,Thomas E. Dever,Christian Kubisch,Lina Basel-Vanagaite +24 more
TL;DR: It is shown that a human X-chromosomal neurological disorder characterized by intellectual disability and microcephaly is caused by a missense mutation in eIF2γ (encoded by EIF2S3), the core subunit of the heterotrimeric eIF3 complex, and this findings directly link intellectual disability to impaired translation initiation.
Journal ArticleDOI
Amino acid substrates impose polyamine, eIF5A, or hypusine requirement for peptide synthesis.
TL;DR: It is proposed that the body of eif5A functionally substitutes for polyamines to promote general protein synthesis and that the hypusine modification on eIF5A is critically important for poor substrates like proline.
Journal ArticleDOI
Structural basis for the transition from translation initiation to elongation by an 80S-eIF5B complex
Jinfan Wang,Jing W. Wang,Byung-Sik Shin,Joo-Ran Kim,Thomas E. Dever,Joseph D. Puglisi,Israel S. Fernández +6 more
TL;DR: The cryoEM structure of the naturally long-lived ribosome complex with eIF5B and Met-tRNAiMet immediately before transition into elongation is determined and uncovers an unexpected, eukaryotic specific and dynamic fidelity checkpoint implemented by eif5B in concert with components of the large ribosomal subunit.
Journal ArticleDOI
Efficacy and safety of cobimetinib (cobi) and atezolizumab (atezo) in an expanded phase 1b study of microsatellite-stable (MSS) metastatic colorectal cancer (mCRC)
Jayesh Desai,Y.S. Hong,Joo-Ran Kim,Tae Won Kim,S.-W. Han,Y-J. Bang,Cheng Ean Chee,V.Y.M. Heong,Autumn J. McRee,Laura Q.M. Chow,E.L. Kwak,J. R. Infante,Jeffrey Wallin,M. Das Thakur,G. Mwawasi,Paul Foster,Edward Cha,Johanna C. Bendell +17 more
