An organoid platform for ovarian cancer captures intra- and interpatient heterogeneity
Oded Kopper,Chris J. de Witte,Kadi Lõhmussaar,Jose Espejo Valle-Inclan,Nizar Hami,Lennart Kester,Anjali Vanita Balgobind,Jeroen Korving,Natalie Proost,Harry Begthel,Lise M van Wijk,Sonia Aristín Revilla,Rebecca Theeuwsen,Marieke van de Ven,Markus J. van Roosmalen,Bas Ponsioen,Victor W.H. Ho,Benjamin G. Neel,Tjalling Bosse,Katja N. Gaarenstroom,Harry Vrieling,Maaike P.G. Vreeswijk,Paul J. van Diest,Petronella O. Witteveen,Trudy N. Jonges,Johannes L. Bos,Alexander van Oudenaarden,Ronald P. Zweemer,Hugo J. Snippert,Wigard P. Kloosterman,Hans Clevers +30 more
TL;DR: A biobank of ovarian cancer organoids recapitulates the histopathological and molecular hallmarks of patient tumors and provides a resource for preclinical research and demonstrates their potential application for research and personalized medicine.
Journal ArticleDOI
Oral Mucosal Organoids as a Potential Platform for Personalized Cancer Therapy
Else Driehuis,S. Kolders,Sacha Spelier,Kadi Lõhmussaar,Stefan M. Willems,Lot A. Devriese,Remco de Bree,Emma J de Ruiter,Jeroen Korving,Harry Begthel,Johan H. van Es,Veerle Geurts,Gui-Wei He,Richard H. van Jaarsveld,Rurika Oka,Mauro J. Muraro,Judith Vivié,Maurice M.J.M. Zandvliet,Antoni P. A. Hendrickx,Nino Iakobachvili,Priya Sridevi,Onno Kranenburg,Ruben van Boxtel,Geert J. P. L. Kops,David A. Tuveson,Peter J. Peters,Alexander van Oudenaarden,Hans Clevers +27 more
TL;DR: Conditions for long-term culture of human mucosal organoids derived from head and neck squamous cell carcinoma (HNSCC) are described and responses to therapies both currently used in the treatment of HNSCC and those not used in clinical practice are observed.
Journal ArticleDOI
Patient-Derived Ovarian Cancer Organoids Mimic Clinical Response and Exhibit Heterogeneous Inter- and Intrapatient Drug Responses.
Chris J. de Witte,Jose Espejo Valle-Inclan,Nizar Hami,Kadi Lõhmussaar,Oded Kopper,Celien P.H. Vreuls,Geertruida N. Jonges,Paul J. van Diest,Luan Nguyen,Hans Clevers,Wigard P. Kloosterman,Edwin Cuppen,Hugo J. Snippert,Ronald P. Zweemer,Petronella O. Witteveen,Ellen Stelloo +15 more
TL;DR: Evaluating the capacity of patient-derived organoids (PDOs) to predict clinical drug response and functional consequences of tumor heterogeneity finds that generating PDOs of multiple tumor locations can improve clinical decision making and increase the knowledge of genetic and drug response heterogeneity.
Journal ArticleDOI
Assessing the origin of high-grade serous ovarian cancer using CRISPR-modification of mouse organoids.
Kadi Lõhmussaar,Oded Kopper,Jeroen Korving,Harry Begthel,Celien P.H. Vreuls,Johan H. van Es,Hans Clevers +6 more
TL;DR: The results support the dual origin hypothesis of HG-SOC, as it is demonstrated that both epithelia can give rise to ovarian tumors with high-grade pathology, however, the mutated oviductal organoids expand much faster in vitro and more readily form malignant tumors upon transplantation.
Journal ArticleDOI
Patient-derived organoids model cervical tissue dynamics and viral oncogenesis in cervical cancer.
Kadi Lõhmussaar,Rurika Oka,Jose Espejo Valle-Inclan,Milou H.H. Smits,Hila Wardak,Jeroen Korving,Harry Begthel,Natalie Proost,Marieke van de Ven,Onno Kranenburg,Trudy N. Jonges,Ronald P. Zweemer,Sebastiaan Veersema,Ruben van Boxtel,Hans Clevers +14 more
TL;DR: In this paper, a long-term culturing protocol for ecto-and endocervical epithelia was described that generates 3D organoids that stably recapitulate the two tissues of origin.
