Peptidomimetic Antibiotics Target Outer-Membrane Biogenesis in Pseudomonas aeruginosa
Nityakalyani Srinivas,Peter Jetter,Bernhard J. Ueberbacher,Martina Werneburg,Katja Zerbe,Jessica Steinmann,Benjamin Van der Meijden,Francesca Bernardini,Alexander Lederer,Ricardo Augusto Dias,Pauline E. Misson,Heiko Henze,Jürg Zumbrunn,Frank Gombert,Daniel Obrecht,Peter Hunziker,Stefan Schauer,Urs Ziegler,Andres Käch,Leo Eberl,Kathrin Riedel,Steven J. Demarco,John A. Robinson +22 more
TL;DR: A family of peptidomimetic antibiotics based on the antimicrobial peptide protegrin I showed potent antimicrobial activity in a mouse septicemia infection model, and may have important therapeutic applications.
Journal ArticleDOI
Chimeric peptidomimetic antibiotics against Gram-negative bacteria
Anatol Luther,Matthias Urfer,Michael Zahn,Maik Müller,Shuang-Yan Wang,Milon Mondal,Alessandra Vitale,Jean-Baptiste Hartmann,Timothy Sharpe,Fabio Lo Monte,Harsha Kocherla,Elizabeth Cline,Gabriella Pessi,Parthasarathi Rath,Seyed Majed Modaresi,Petra Chiquet,Sarah Stiegeler,Carolin Verbree,Tobias Remus,Michel Schmitt,Caroline Kolopp,Marie-Anne Westwood,Nicolas Desjonquères,Emile Brabet,Sophie Hell,Karen LePoupon,Annie Vermeulen,Régis Jaisson,Virginie Rithié,Gregory Upert,Alexander Lederer,Peter Zbinden,Achim Wach,Kerstin Moehle,Katja Zerbe,Hans H. Locher,Francesca Bernardini,Glenn E. Dale,Leo Eberl,Bernd Wollscheid,Sebastian Hiller,John A. Robinson,Daniel Obrecht +42 more
TL;DR: A class of chimeric synthetic antibiotics that bind to lipopolysaccharide and BamA shows potent activity against multidrug-resistant Gram-negative bacteria, with the potential to address life-threatening infections.
Journal ArticleDOI
Crystal Structure of OxyB, a Cytochrome P450 Implicated in an Oxidative Phenol Coupling Reaction during Vancomycin Biosynthesis
Katja Zerbe,Olena Pylypenko,Francesca Vitali,Weiwen Zhang,Severine Rouset,Markus Heck,Jan Wim Vrijbloed,Daniel Bischoff,Bojan Bister,Roderich D. Süssmuth,Stefan Pelzer,Wolfgang Wohlleben,John A. Robinson,Ilme Schlichting +13 more
TL;DR: Gene-inactivation studies point to the involvement of OxyB in catalyzing the first oxidative phenol coupling reaction during glycopeptide antibiotic biosynthesis, and OxyB exhibits the typical P450-fold.
Journal ArticleDOI
An Oxidative Phenol Coupling Reaction Catalyzed by OxyB, a Cytochrome P450 from the Vancomycin-Producing Microorganism†
Journal ArticleDOI
Oxidative phenol coupling reactions catalyzed by OxyB: a cytochrome P450 from the vancomycin producing organism. implications for vancomycin biosynthesis.
Katharina Woithe,Nina Geib,Katja Zerbe,Dong Bo Li,Markus Heck,Severine Fournier-Rousset,Odile Meyer,Francesca Vitali,Nobuatsu Matoba,Khaled Abou-Hadeed,John A. Robinson +10 more
TL;DR: It is shown that cross-linking of Hpg4 and Tyr6 by OxyB can occur in both hexapeptide- and heptapeptid-PCP conjugates, and it is possible that a linear heptAPEptide intermediate linked to PCP-7 in NRPS subunit-3 may also be transformed into monocyclic product.
