TNF-α Mediates PKR-Dependent Memory Impairment and Brain IRS-1 Inhibition Induced by Alzheimer’s β-Amyloid Oligomers in Mice and Monkeys
Mychael V. Lourenco,Julia R. Clarke,Rudimar Luiz Frozza,Theresa R. Bomfim,Leticia Forny-Germano,Andre F. Batista,Luciana B. Sathler,Jordano Brito-Moreira,Olavo B. Amaral,Cesar A. Silva,Léo Freitas-Correa,Sheila Espírito-Santo,Paula Campello-Costa,Jean-Christophe Houzel,William L. Klein,Christian Hölscher,José B.C. Carvalheira,Aristóbolo M. Silva,Aristóbolo M. Silva,Licio A. Velloso,Douglas P. Munoz,Sergio T. Ferreira,Fernanda G. De Felice +22 more
TL;DR: Results reveal pathogenic mechanisms shared by AD and diabetes and establish that proinflammatory signaling mediates oligomer-induced IRS-1 inhibition and PKR-dependent synapse and memory loss.
Journal ArticleDOI
Amyloid-β Oligomers Induce Differential Gene Expression in Adult Human Brain Slices
Adriano Sebollela,Léo Freitas-Correa,Fabio F. Oliveira,Andrea C. Paula-Lima,Leonardo M. Saraiva,Samantha M. Martins,Louise Danielle de Carvalho Mota,Cesar Torres,Soniza Vieira Alves-Leon,Jorge Marcondes de Souza,Dirce Maria Carraro,Helena Brentani,Fernanda G. De Felice,Sergio T. Ferreira +13 more
TL;DR: Investigation of changes in transcription in adult human cortical slices exposed to sublethal doses of AβOs establishes early mechanisms involved in AβO-triggered neuronal dysfunction in a novel human-derived experimental model and suggests that the neuronal pathways affected by A βOs may be targets for the development of novel diagnostic or therapeutic approaches.
Journal ArticleDOI
Expression Profile of Rat Hippocampal Neurons Treated with the Neuroprotective Compound 2,4-Dinitrophenol: Up-Regulation of cAMP Signaling Genes
Adriano Sebollela,Léo Freitas-Correa,Fabio F. Oliveira,Camila T. Mendes,Ana Paula Wasilewska-Sampaio,Juliana Camacho-Pereira,Antonio Galina,Helena Brentani,Fabio Passetti,Fernanda G. De Felice,Emmanuel Dias-Neto,Sergio T. Ferreira +11 more
TL;DR: DNA microarray analysis is used to investigate changes in gene expression in rat hippocampal neurons in culture treated with low micromolar concentrations of DNP, shedding light on molecular mechanisms underlying neuroprotection by DNP and point to possible targets for development of novel therapeutics for neurodegenerative disorders.
Journal ArticleDOI
2,4-dinitrophenol induces neural differentiation of murine embryonic stem cells
Léo Freitas-Correa,Mychael V. Lourenco,Mariana Acquarone,Rodrigo Madeiro da Costa,Antonio Galina,Stevens K. Rehen,Sergio T. Ferreira +6 more
TL;DR: It is established that D NP induces neural differentiation of ESCs, accompanied by cell proliferation, migration and neuritogenesis, suggesting that DNP may be a novel tool to induce neurogenesis in embryonic stem cells.
