Oncogenic IL7R gain-of-function mutations in childhood T-cell acute lymphoblastic leukemia.
Priscila Pini Zenatti,Daniel Dias Ribeiro,Wenqing Li,Linda Zuurbier,Milene Costa da Silva,Maddalena Paganin,Julia Tritapoe,Julie A. Hixon,André B. Silveira,Bruno A. Cardoso,Leonor M. Sarmento,Nádia C. Correia,María L. Toribio,Joerg Kobarg,Martin A. Horstmann,Rob Pieters,Silvia Regina Brandalise,Adolfo A. Ferrando,Jules P.P. Meijerink,Scott K. Durum,J. Andrés Yunes,João T. Barata +21 more
TL;DR: The findings indicate that IL7R mutational activation is involved in human T-cell leukemogenesis, paving the way for therapeutic targeting of IL-7R–mediated signaling in T-ALL.
Journal ArticleDOI
PTEN microdeletions in T-cell acute lymphoblastic leukemia are caused by illegitimate RAG-mediated recombination events
Rui D. Mendes,Leonor M. Sarmento,Kirsten Canté-Barrett,Linda Zuurbier,Jessica G.C.A.M. Buijs-Gladdines,Vanda Póvoa,Willem K. Smits,Miguel Abecasis,J. Andrés Yunes,Edwin Sonneveld,Martin A. Horstmann,Rob Pieters,João T. Barata,Jules P.P. Meijerink +13 more
TL;DR: It is shown that PTEN is inactivated by small deletions affecting a few exons in 8% of pediatric T-ALL patients, and ongoing RAG activity may actively contribute to the acquisition of preleukemic hits, clonal diversification, and disease progression.
Journal ArticleDOI
CHK1 overexpression in T-cell acute lymphoblastic leukemia is essential for proliferation and survival by preventing excessive replication stress
Leonor M. Sarmento,Vanda Póvoa,R. Nascimento,Gonçalo Real,I Antunes,Leila R. Martins,Catarina Moita,Paula M. Alves,Miguel Abecasis,Luis F. Moita,R. M. E. Parkhouse,Jules P.P. Meijerink,João T. Barata +12 more
TL;DR: It is concluded that CHK1 is critical for T-ALL proliferation and viability by downmodulating replication stress and preventing ATM/caspase-3-dependent cell death.
Journal ArticleDOI
microRNAs regulate TAL1 expression in T-cell acute lymphoblastic leukemia
Nádia C. Correia,Alice Melão,Vanda Póvoa,Leonor M. Sarmento,Marta Gómez de Cedrón,Marcos Malumbres,Francisco J. Enguita,João T. Barata +7 more
TL;DR: The results show for the first time the existence of epigenetic regulation of TAL1 by specific miRNAs which may contribute, at least in part, to the ectopic expression of Tal1 in some T-ALL cases.
Journal ArticleDOI
Therapeutic potential of Notch inhibition in T-cell acute lymphoblastic leukemia: rationale, caveats and promises
TL;DR: The latest advances in the development and use of Notch antagonists are reviewed and the current knowledge on Notch function in T-ALL is summarized to understand how it may translate into novel therapeutic strategies that increment the efficiency of notch inhibition.
