Journal ArticleDOI
Structural Insight into Eukaryotic Sterol Transport through Niemann-Pick Type C Proteins.
Mikael B. L. Winkler,Rune T. Kidmose,Maria Szomek,Katja Thaysen,Shaun Rawson,Stephen P. Muench,Daniel Wüstner,Bjørn Panyella Pedersen +7 more
TL;DR: A model for sterol integration is proposed that clarifies the role of NPC proteins in this essential eukaryotic pathway and that rationalizes mutations in patients with Niemann-Pick disease type C.
Journal ArticleDOI
Ergosterol is mainly located in the cytoplasmic leaflet of the yeast plasma membrane
Lukasz M. Solanko,David P. Sullivan,Yves Y. Sere,Maria Szomek,Anita Lunding,Katarzyna A. Solanko,Azra Pizovic,Lyubomir Dimitrov Stanchev,Lyubomir Dimitrov Stanchev,Thomas Günther Pomorski,Thomas Günther Pomorski,Anant K. Menon,Daniel Wüstner +12 more
TL;DR: It is deduced that sterols comprise up to ~45% of all inner leaflet lipids in the PM, a result that necessitates revision of current models of the architecture of the PM lipid bilayer.
Journal ArticleDOI
Niemann-Pick C2 protein regulates sterol transport between plasma membrane and late endosomes in human fibroblasts
Zane Berzina,Lukasz M. Solanko,Ahmed S Mehadi,Maria Louise V. Jensen,Frederik W. Lund,Maciej Modzel,Maria Szomek,Katarzyna A. Solanko,Alice Dupont,Gitte Nielsen,Christian W. Heegaard,Christer S. Ejsing,Daniel Wüstner +12 more
TL;DR: This study shows that cholesterol esterification and trafficking of sterols between the PM and LE/LYSs depends on a functional NPC2 protein, which likely acts inside LE/LySs from where it increases non-vesicular sterol exchange with other organelles.
Journal ArticleDOI
Niemann Pick C2 protein enables cholesterol transfer from endo-lysosomes to the plasma membrane for efflux by shedding of extracellular vesicles.
Alice Dupont Juhl,Frederik W. Lund,Maria Louise V. Jensen,Maria Szomek,Christian W. Heegaard,Peter Guttmann,Stephan Werner,James G. McNally,Gerd Schneider,Sergey Kapishnikov,Daniel Wüstner +10 more
TL;DR: In this paper, the Niemann-Pick C2 protein (NPC2) was found to facilitate non-vesicular sterol exchange with the plasma membrane for subsequent sterol efflux to acceptor proteins and shedding of sterol-rich vesicles from the cell surface.
Journal ArticleDOI
Substituted 9-Diethylaminobenzo[a]phenoxazin-5-ones (Nile Red Analogues): Synthesis and Photophysical Properties.
Mick Hornum,Mads W. Mulberg,Maria Szomek,Peter Reinholdt,Jonathan R. Brewer,Daniel Wüstner,Jacob Kongsted,Poul Nielsen +7 more
TL;DR: In this article, the authors present synthesis approaches to a series of monosubstituted Nile Red derivatives (9-diethylbenzo[a]phenoxazin-5-ones) starting from 1-naphthols or 1,3naphthalenediols.
