Journal ArticleDOI
Clonal Hematopoiesis and risk of atherosclerotic cardiovascular disease
Siddhartha Jaiswal,Pradeep Natarajan,Pradeep Natarajan,Alexander J. Silver,Christopher J. Gibson,Alexander G. Bick,Eugenia Shvartz,Marie McConkey,Namrata Gupta,Stacey Gabriel,Diego Ardissino,Usman Baber,Roxana Mehran,Valentin Fuster,Valentin Fuster,John Danesh,Philippe M. Frossard,Danish Saleheen,Olle Melander,Olle Melander,Galina K. Sukhova,Donna Neuberg,Peter Libby,Sekar Kathiresan,Benjamin L. Ebert +24 more
TL;DR: The presence of CHIP in peripheral‐blood cells was associated with nearly a doubling in the risk of coronary heart disease in humans and with accelerated atherosclerosis in mice.
Journal ArticleDOI
Lenalidomide Causes Selective Degradation of IKZF1 and IKZF3 in Multiple Myeloma Cells
Jan Krönke,Namrata D. Udeshi,Anupama Narla,Peter V. Grauman,Slater N. Hurst,Marie McConkey,Tanya Svinkina,Dirk Heckl,Eamon Comer,Xiaoyu Li,Christie Ciarlo,Emily C. Hartman,Nikhil C. Munshi,Monica Schenone,Stuart L. Schreiber,Steven A. Carr,Benjamin L. Ebert,Benjamin L. Ebert +17 more
TL;DR: Using quantitative proteomics, it is found that lenalidomide causes selective ubiquitination and degradation of two lymphoid transcription factors, IKZF1 and IKzF3, by the CRBN-CRL4 ubiquitin ligase, which are essential transcription factors in multiple myeloma.
Journal ArticleDOI
Lenalidomide induces ubiquitination and degradation of CK1α in del(5q) MDS
Jan Krönke,Emma C. Fink,Emma C. Fink,Paul W. Hollenbach,Kyle J. MacBeth,Slater N. Hurst,Namrata D. Udeshi,Philip P Chamberlain,D. R. Mani,Hon-Wah Man,Anita Gandhi,Tanya Svinkina,Rebekka K. Schneider,Marie McConkey,Marcus Järås,Elizabeth A. Griffiths,Meir Wetzler,Lars Bullinger,Brian E. Cathers,Steven A. Carr,Rajesh Chopra,Benjamin L. Ebert,Benjamin L. Ebert +22 more
TL;DR: It is demonstrated that lenalidomide induces the ubiquitination of casein kinase 1A1 (CK1α) by the E3 ubiquitin ligase CUL4–RBX1–DDB1–CRBN (known as CRL4CRBN), resulting in CK1α degradation.
Journal ArticleDOI
Physiologic Expression of Sf3b1(K700E) Causes Impaired Erythropoiesis, Aberrant Splicing, and Sensitivity to Therapeutic Spliceosome Modulation.
Esther A. Obeng,Esther A. Obeng,Esther A. Obeng,Ryan J. Chappell,Michael Seiler,Michelle C. Chen,Dean R. Campagna,Paul J. Schmidt,Rebekka K. Schneider,Allegra M. Lord,Lili Wang,Rutendo Gambe,Marie McConkey,Abdullah Mahmood Ali,Azra Raza,Lihua Yu,Silvia Buonamici,Pete Smith,Ann Mullally,Ann Mullally,Catherine J. Wu,Catherine J. Wu,Mark D. Fleming,Benjamin L. Ebert,Benjamin L. Ebert +24 more
TL;DR: SF3B1(K700E) expression reflects the phenotype of the mutation in myelodysplastic syndrome and may be a therapeutic target in MDS.
Journal ArticleDOI
Increased neutrophil extracellular trap formation promotes thrombosis in myeloproliferative neoplasms.
Ofir Wolach,Ofir Wolach,Ofir Wolach,Rob S. Sellar,Rob S. Sellar,Rob S. Sellar,Kimberly Martinod,Deya Cherpokova,Marie McConkey,Ryan J. Chappell,Alexander J. Silver,Dylan N. Adams,Cecilia A. McCarroll,Steven A. McCarroll,Bozenna Iliadou,Christina M. Hultman,Donna Neuberg,Ann Mullally,Ann Mullally,Ann Mullally,Denisa D. Wagner,Benjamin L. Ebert,Benjamin L. Ebert,Benjamin L. Ebert +34 more
TL;DR: It is demonstrated that neutrophils from patients with MPNs are primed for NET formation, an effect blunted by pharmacological inhibition of JAK signaling, and suggested that JAK2 inhibition may reduce thrombosis in MPNs through cell-intrinsic effects on neutrophil function.
