Journal ArticleDOI
Phosphatidylinositol 4,5-bisphosphate induces actin-based movement of raft-enriched vesicles through WASP-Arp2/3
Andrew L. Rozelle,Laura M. Machesky,Masaya Yamamoto,Mariëtte H.E. Driessens,Robert H. Insall,Michael G. Roth,Katherine Luby-Phelps,Gerard Marriott,Alan Hall,Helen L. Yin +9 more
TL;DR: Overexpression of type I phosphatidylinositol phosphate 5-kinase (PIP5KI), which synthesizes PIP(2), promoted actin polymerization from membrane-bound vesicles to form motile actin comets, establishing that rafts promote comet formation.
Journal ArticleDOI
Cdc42 induces filopodia by promoting the formation of an IRSp53:Mena complex.
Sonja Krugmann,Ingrid Jordens,Kris Gevaert,Mariëtte H.E. Driessens,Joël Vandekerckhove,Alan Hall +5 more
TL;DR: It is concluded that the interaction of Cdc42 with the partial CRIB motif of IRSp53 relieves an intramolecular, autoinhibitory interaction with the N terminus, allowing the recruitment of Mena to theIRSp53 SH3 domain.
Journal ArticleDOI
Plexin-B semaphorin receptors interact directly with active Rac and regulate the actin cytoskeleton by activating Rho
Mariëtte H.E. Driessens,Hailan Hu,Catherine D. Nobes,Annette J. Self,Ingrid Jordens,Corey S. Goodman,Alan Hall +6 more
TL;DR: The findings that plexin-B signaling to the cytoskeleton is both Rac and Rho dependent form a starting point for unraveling the mechanism by which semaphorins and plexins control axon guidance and cell migration.
Journal ArticleDOI
Application of SHAP values for inferring the optimal functional form of covariates in pharmacokinetic modeling
Mark Hoogendoorn,Marjon H. Cnossen,S. H. Reitsma,Frank W.G. Leebeek,Ron A. A. Mathôt,Karin Fijnvandraat,Michiel Coppens,Karina Meijer,Saskia E M Schols,H. C. J. B. de Jager,Laura H. Bukkems,Michael E. Cloesmeijer,A. Janssen,Patrick William Collins,Ri Liesner,Pavani Chowdary,C M Millar,D Hart,Desmond Keeling +39 more
TL;DR: It is concluded that the proposed method can be used to extend ML‐based covariate selection, and holds potential as a complete full model alternative to classical covariate analyses.
Journal ArticleDOI
Deep compartment models: A deep learning approach for the reliable prediction of time‐series data in pharmacokinetic modeling
Alexander Janssen,Frank W.G. Leebeek,Marjon H. Cnossen,Ron A. A. Mathôt,Karin Fijnvandraat,Michiel Coppens,Karina Meijer,Saskia E M Schols,H. C. J. Eikenboom,Reg Schutgens,Erik A M Beckers,Paula F. Driessens,Iris Van de Vliet,Janske Lock,Hendrika C A M Hazendonk,Iris van Moort,Jessica M. Heijdra,Marc Goedhart,Wala Al Arashi,Tim Preijers,Nico C. B. de Jager,Laura H. Bukkems,Michael E. Cloesmeijer,Patrick William Collins,Ri Liesner,Pavani Chowdary,C M Millar,D Hart,Desmond Keeling +36 more
TL;DR: The deep compartment model (DCM), a combination of neural networks and ordinary differential equations, is proposed and it is shown that the model remains accurate when training on small data sets and correctly describes the changing drug concentration over time.
