Evidence from clinical and experimental studies indicates that macrophages promote solid-tumour progression and metastasis. Macrophages are educated by the tumour microenvironment, so that they adopt a trophic role that facilitates angiogenesis, matrix breakdown and tumour-cell motility — all of which are elements of the metastatic process. During an inflammatory response, macrophages also produce many compounds — ranging from mutagenic oxygen and nitrogen radicals to angiogenic factors — that can contribute to cancer initiation and promotion. Macrophages therefore represent an important drug target for cancer prevention and cure.

Personalizing the treatment of women with early breast cancer: highlights of the St Gallen International Expert Consensus on the Primary Therapy of Early Breast Cancer 2013

Aim: To evaluate the antiemetic efficacy and safety of olanzapine in Japanese women with nausea and/or vomiting refractory to the standard antiemetic regimen. Methods: We retrospectively reviewed the medical records of consecutive female patients with early breast cancer who underwent highly emetogenic chemotherapy at our hospital from January 2009 to March 2013. Patients with grade 2 or 3 nausea and/or vomiting despite receiving standard antiemetics (5-hydroxytryptamine3 receptor antagonists and dexamethasone 20 mg on day 1, and 4 mg dexamethasone on days 2 and 3) in the first chemotherapy cycle received an additional 2.5–10 mg olanzapine from days 1 to 3 in the subsequent cycles. We assessed patient characteristics, olanzapine dose, nausea and vomiting grades before and after adding olanzapine, and adverse effects. Results: We reviewed 20 patients with poor control in the first chemotherapy cycle, despite receiving standard antiemetics, who received olanzapine from the second cycle. Tumor immune microenvironment and systemic response in breast cancer

We have investigated the change of balance between proliferating and apoptotic tumor cells in dimethylebenz(a)-anthracene (DMBA)-induced rat mammary tumors by using anti-angiogenic drug, AGM-1470 and anticancer drug, 5'-deoxy-5-fluorouridine (5'-DFUR). The rate of PCNA positive proliferating cells was decreased in the tretment groups, especially in AGM-1470 and in the combination of AGM-1470 and 5'-DFUR. And apoptotic index evaluated by TUNEL method was increased in 5'-DFUR and in the combination group. The ratio of proliferating cells/apoptotic cells was markedly decreased in AGM-1470 and AGM+5'-DFUR group compared to control group. These data suggested that anti-angiogenic drugs might regulate the growth behavior of tumor cells by changing the microenvironments.
