A microfluidic culture platform for CNS axonal injury, regeneration and transport
Anne Marion Taylor,Mathew Blurton-Jones,Seog Woo Rhee,David H. Cribbs,Carl W. Cotman,Neon Li Jeon +5 more
TL;DR: A microfluidic culture platform that polarizes the growth of CNS axons into a fluidically isolated environment without the use of targeting neurotrophins is described and the first evidence that presynaptic but not postsynaptic mRNA is localized to developing rat cortical and hippocampal axons is reported.
Journal ArticleDOI
Neural stem cells improve cognition via BDNF in a transgenic model of Alzheimer disease.
Mathew Blurton-Jones,Masashi Kitazawa,Hilda Martinez-Coria,Nicholas A. Castello,Franz-Josef Müller,Jeanne F. Loring,Tritia R. Yamasaki,Wayne W. Poon,Kim N. Green,Frank M. LaFerla +9 more
TL;DR: Interestingly, despite widespread and established Aß plaque and neurofibrillary tangle pathology, hippocampal neural stem cell transplantation rescues the spatial learning and memory deficits in aged 3xTg-AD mice.
Journal ArticleDOI
iPSC-Derived Human Microglia-like Cells to Study Neurological Diseases
Edsel M. Abud,Ricardo Ramirez,Eric S. Martinez,Luke M. Healy,Cecilia H.H. Nguyen,Sean A. Newman,Andriy V. Yeromin,Vanessa M. Scarfone,Samuel E. Marsh,Cristhian Fimbres,Chad A. Caraway,Gianna M. Fote,Abdullah M. Madany,Anshu Agrawal,Rakez Kayed,Karen H. Gylys,Michael D. Cahalan,Brian J. Cummings,Jack P. Antel,Ali Mortazavi,Monica J. Carson,Wayne W. Poon,Mathew Blurton-Jones +22 more
TL;DR: iMGLs were used to examine the effects of Aβ fibrils and brain-derived tau oligomers on AD-related gene expression and to interrogate mechanisms involved in synaptic pruning, and whole-transcriptome analysis demonstrates that they are highly similar to cultured adult and fetal human microglia.
Journal ArticleDOI
Caspase-cleavage of tau is an early event in Alzheimer disease tangle pathology
Robert A. Rissman,Wayne W. Poon,Mathew Blurton-Jones,Salvatore Oddo,Reidun Torp,Michael P. Vitek,Frank M. LaFerla,Troy T. Rohn,Carl W. Cotman +8 more
TL;DR: It is demonstrated that tau is cleaved at D421 (DeltaTau) by executioner caspases, and therapeutics aimed at inhibiting tau caspase-cleavage may prove beneficial not only in preventing NFT formation, but also in slowing cognitive decline.
Journal ArticleDOI
Eliminating microglia in Alzheimer’s mice prevents neuronal loss without modulating amyloid-β pathology
Elizabeth E. Spangenberg,Rafael J. Lee,Allison R. Najafi,Rachel A. Rice,Monica R. P. Elmore,Mathew Blurton-Jones,Brian L. West,Kim N. Green +7 more
TL;DR: Chronic microglial elimination does not alter amyloid-β levels or plaque load; however, it does rescue dendritic spine loss and prevent neuronal loss in 5xfAD mice, as well as reduce overall neuroinflammation.
