Oncogenic PI3K mutations are as common as AKT1 and SMO mutations in meningioma.
Malak Abedalthagafi,Wenya Linda Bi,Ayal A. Aizer,Parker H. Merrill,Ryan Brewster,Pankaj K. Agarwalla,Marc L. Listewnik,Dora Dias-Santagata,Aaron R. Thorner,Paul Van Hummelen,Priscilla K. Brastianos,David A. Reardon,Patrick Y. Wen,Ossama Al-Mefty,Shakti Ramkissoon,Rebecca D. Folkerth,Keith L. Ligon,Azra H. Ligon,Brian M. Alexander,Ian F. Dunn,Rameen Beroukhim,Sandro Santagata +21 more
TL;DR: This work identifies PI3K signaling as an important target for precision medicine trials in meningioma patients using high-resolution array-comparative genomic hybridization to prospectively characterize copy-number changes.
Journal ArticleDOI
Genomic characterization of human brain metastases identifies drivers of metastatic lung adenocarcinoma
David Shih,David Shih,Naema Nayyar,Ivanna Bihun,Ivanna Bihun,Ibiayi Dagogo-Jack,Corey M. Gill,Corey M. Gill,Elisa Aquilanti,Elisa Aquilanti,Mia Bertalan,Alexander Kaplan,Megan R. Brastianos +51 more
TL;DR: It is demonstrated that somatic alterations contribute to brain metastases and that genomic sequencing of a sufficient number of metastatic tumors can reveal previously unknown metastatic drivers.
Journal ArticleDOI
Germline and somatic BAP1 mutations in high-grade rhabdoid meningiomas
Ganesh M. Shankar,Malak Abedalthagafi,Rachael A. Vaubel,Parker H. Merrill,Naema Nayyar,Corey M. Gill,Ryan Brewster,Wenya Linda Bi,Pankaj K. Agarwalla,Aaron R. Thorner,David A. Reardon,Ossama Al-Mefty,Patrick Y. Wen,Brian M. Alexander,Paul Van Hummelen,Tracy T. Batchelor,Keith L. Ligon,Azra H. Ligon,Matthew Meyerson,Ian F. Dunn,Rameen Beroukhim,David N. Louis,Arie Perry,Scott L. Carter,Caterina Giannini,William T. Curry,Daniel P. Cahill,Fred G. Barker,Priscilla K. Brastianos,Sandro Santagata +29 more
TL;DR: It is shown that familial and sporadic BAP1-mutated rhabdoid meningiomas are clinically aggressive, requiring intensive clinical management, and implicate ubiquitin deregulation in the pathogenesis of these high-grade malignancies.
Journal ArticleDOI
ARID1A and TERT promoter mutations in dedifferentiated meningioma.
Malak Abedalthagafi,Wenya Linda Bi,Parker H. Merrill,William J. Gibson,Matthew F. Rose,Ziming Du,Joshua M. Francis,Joshua M. Francis,Rose Du,Ian F. Dunn,Azra H. Ligon,Rameen Beroukhim,Rameen Beroukhim,Sandro Santagata,Sandro Santagata +14 more
TL;DR: A case of an intraventricular meningioma that at first presentation displayed remarkable morphologic heterogeneity-composed of distinct regions independently fulfilling histopathologic criteria for WHO grade I, II, and III designations is presented.
Journal ArticleDOI
Compromising the 19S proteasome complex protects cells from reduced flux through the proteasome
Peter Tsvetkov,Marc L. Mendillo,Jinghui Zhao,Jan E. Carette,Parker H. Merrill,Domagoj Cikes,Malini Varadarajan,Ferdy R. van Diemen,Josef M. Penninger,Alfred L. Goldberg,Thijn R. Brummelkamp,Sandro Santagata,Susan Lindquist +12 more
TL;DR: Counter to expectation, reducing expression of individual subunits of the proteasome's 19S regulatory complex increased survival, and this means of rebalancing proteostasis is conserved from yeast to humans.
